摘  要:microRNA(miRNA)是一类长19~24个碱基的小分子内源性非编码单链RNA,通过介导靶基因的裂解或抑制靶基因的翻译,在植物的发育、新陈代谢、应激响应等方面起着重要的调控作用,在进化上具有高度的保守性。据此本研究以miRBase注册的全部成熟植物miRNA序列为探针,基于NCBI数据库中茶树表达序列标签(EST)和基因组勘测序列(GSS)进行同源搜索和miRNA二级结构分析,总共发现7条茶树的miRNA,分属于6个不同的miRNA家族。通过靶基因的预测分析表明,这些miRNA可能调节28种靶标基因,功能涉及新陈代谢、生长发育、胁迫应答及转录,为茶树miRNA的试验鉴定和功能研究提供更详细精确的范围,为进一步探究茶树miRNA的调控机理奠定基础。microRNAs(miRNAs) are approximately 19-24 nucleotides endogenous noncoding single-stranded RNAs produced. Many plant miRNAs, highly conserved, have critical functions in growth, development, metabolism, abiotic stress responses and so on with specific target mRNAs. In this study, we uesed previously deposited miRNA sequence from all mature plant miRNA sequences in miRBase to blast the database of tea plant expressed sequence tags(EST) and genomic survey sequence(GSS) that do not correspond to protein coding genes. Following the homology search and miRNA secondary structure analysis, seven potential miRNAs of six different miRNA families were detected in tea plant. Analysis of those eight new miRNAs via in NCBI showed that they might regulate 28 target genes. These target genes involved in the regulation of processes including metabolism, growth, development, abiotic stress responses and transcriptional regulation. Our reseach provides more detailed and precise ranges for tea miRNA identification and functional studies, laying the foundation for further investigation of the miRNA regulatory mechanisms in tea plant.
