摘 要:目的研究钙离子(Ca^(2+))对针刺神经电信号的影响,为揭示Ca^(2+)在针刺信号传导过程中的作用及针效产生的始动机制进一步提供实验依据。方法选用健康成年雄性SD大鼠,实验组采用0.1 mol/L乙二醇双乙胺醚四乙酸(EGTA)5mL穴位注射络合足三里穴区Ca^(2+),对照组采用0.05 mol/L Ca^(2+)-EGTA对照液5mL穴位注射。分别记录穴位注射前(YQ)及穴位注射后即刻(YH0)、7 min(YH7)、14 min(YH14)、21 min(YH21)、28 min(YH28)时神经细束的放电情况。应用非线性时间序列分析方法、小波能量熵分析法、峰峰间期等方法对采集的数据进行分析。结果实验组各时间段(YH0、YH21、YH28)的放电频率均降低,与同组YQ比较差异均具有统计学意义(P<0.01);对照组各时间段(YH14、YH21、YH28)的放电频率显著增高,与同组YQ比较差异均具有统计学意义(P<0.05)。实验组YH21、YH28的放电频率与对照组比较差异均具有统计学意义(P<0.05)。通过放电频率、波形幅值的统计发现穴区Ca^(2+)浓度降低对针刺引发的放电序列有比较明显的影响。结论 Ca^(2+)是针效产生机制的重要环节之一,其在针刺神经电信号传导过程中起着重要作用。Objective To investigate the effect of calcium ions (Ca^2+) on electroneurographic signals of acupuncture, and to provide experimental evidence to reveal the effect of Ca^2+ on the process of signal transduction of acupuncture and the initial mechanism of acupuncture effect. Method Healthy adult male Sprague-Dawley (SD) rats were selected. The experimental group received acupoint injection of 5 μL ethyleneglycol-bis-(2-aminoethyl ether)-N,N,N',N'- tetraacetic acid (EGTA) (0.1 mol/L) to chelate Ca^2+ in the area of Zusanli (ST36), while the control group received acupoint injection of 5μL Ca^2+-EGTA (0.05 mol/L). The electrical discharge of nerve fiber bundle was respectively recorded before acupoint injection (YQ), immediately after acupoint injection (YH0), and at 7 min (YH7), 14 min (YH14), 21 min (YH21) and 28 min (YH28) after acupoint injection. The collected data were analyzed by nonlinear time series analysis, wavelet energy entropy analysis and interspike intervals. Result In the experimental group, the discharge frequency at different time points (YH0, YH21 and YH28) decreased, and was significantly different from that at YQ (P〈0.01). In the control group, the discharge frequency at different time points (YH14, YH21 and YH28)increased, and was significantly different from that at YQ (P〈0.05). There was a significant difference in the discharge frequency at different time points (YH21 and YH28) between the two groups (P〈0.05). The reduction of Ca^2+ concentration in the area of acupoint had a relatively obvious effect on the discharge sequence caused by acupuncture according to the statistics of the discharge frequency and the waveform amplitude. Conclusion Ca^2+ is one of the important links in the mechanism of acupuncture effect, as it plays an important role in the process of electroneuro- graphic signal transduction of acupuncture.
