摘 要:目的研究TLR4 rs1927907、rs11536889基因多态性与慢性牙周炎及COPD下游炎性通路中细胞因子的关系。方法选择慢性牙周炎患者81例,COPD患者27例,慢性牙周炎伴COPD患者76例,健康对照者26例。采集所有受试对象的静脉血,分离血细胞与血清。提取白细胞基因组DNA,采用TaqMan MGB探针法对TLR4 rs1927907、rs11536889进行基因型检测。采用酶联免疫吸附法检测血清中IL-1β、IL-6、TNF-α的水平。结果牙周炎组,rs11536889 GG基因型患者血清中的TNF-α水平显著低于GC+CC基因型患者(P=0.038)。慢性牙周炎伴COPD组中,rs11536889 GG基因型患者血清中的IL-6水平显著高于GC+CC基因型患者(P=0.006)。其余2组rs1927907、rs11536889基因型与血清中IL-1β、IL-6、TNF-α水平无显著差异(P>0.05)。结论在慢性牙周炎组和慢性牙周炎伴COPD组中,TLR4 rs11536889基因多态性与TNF-α、IL-6的表达有关,可能增强了机体介导的炎症反应。Objective To investigate the association of TLR4 rs1927907, rs11536889 gene polymorphisms and cytokine between chronic periodontitis and COPD. Methods 81 patients with chronic periodontitis, 27 patients with COPD, 76 patients with chronic periodontitis with COPD, and 26 healthy controls were collected. Venous blood from all subjects was taken. Blood cells and serum were separated. Leukocyte genomic DNA was extracted, and TLR4 rs1927907 and rs11536889 were genotyped by TaqMan MGB probe method. The levels of IL-1β, IL-6 and TNF-α in serum were detected by enzyme linked immunosorbent assay. Results In the periodontitis group, the TNF-α level in the serum of patients with rs11536889 GG genotype was significantly lower than that of patients with GC+CC genotype (P =0.038). In the chronic periodontitis with COPD group, IL-6 levels in the serum of patients with rs11536889 GG genotype were significantly higher than those with GC+CC genotype (P =0.006). There were no significant differences in the levels of IL-1β, IL-6 and TNF-α between the other two groups of rs1927907 and rs11536889 genotypes(P 〉0.05). Conclusion The TLR4 rs11536889 genotype polymorphism is associated with the expression of TNF-α and IL-6 in chronic periodontitis group and chronic periodontitis with COPD group, which may enhance the inflammatory response.
