摘 要:猪的生长性状是重要的经济性状之一,直接影响到养猪企业的经济效益,同时还间接反映了猪育种工作的现状和进展。为了了解猪生长性状的遗传机制,寻找与生长性状显著相关的位点,进而定位与生长性状有关联的功能基因,研究利用全基因组关联分析(Genome-wide Association Study, GWAS)研究猪的生长性状相关基因。试验选择571头美系大白猪作为研究对象,采用基于线性模型的单标记回归方法,同时借助Illumina猪80 K高密度SNP芯片(共有68 528个SNP位点)对样本个体的基因型进行分型,对大白猪的两个生长性状100 kg体重活体的背膘厚和达100 kg体重的日龄,进行了全基因组关联分析。研究将群体进行全基因组关联分析后使用FDR方法对关联分析的结果进行多重检验校正,发现了4个位于15号染色体上与100 kg体重日龄性状相关的基因组水平显著的SNP位点,分别是WU_10.2_15_142776834、WU_10.2_15_142743420、WU_10.2_15_142698412、WU_10.2_15_142730007。2个位于1号染色体上与活体背膘厚性状相关的基因组水平显著的SNP位点,分别是ALGA0005769、ASGA0004384。进而利用EnseMble在线平台的猪SusScrofa10.2版本数据库,将显著的SNP位点向上下游各扩展1 Mb进行基因富集分析,定位到了可能与生长性状有关联的基因。其中,与100 kg体重日龄相关的基因有9个,分别是SLC19A3、CCL20、DAW1、SPHKAP、RHBDD1、IRS1、COL4A4、TM4SF20、AGFG1;与活体背膘厚相关的基因有16个,分别是ONECUT1、ARPP19、MYO5A、MYO5C、GNB5、BCL2L10、MAPK6、LEO1、TMOD3、SCG3、LYSMD2、TMOD2、DMXL2、GLDN、CYP19A1、TNFAIP8L3。结论,ARPP19可以控制有丝分裂,ONEUT1具有促进细胞分裂和生长发育的功能。因此这两个基因最可能与生长性状存在功能联系。Growth traits of pigs are important economic traits, these traits not only aftect the economic performance of the pig business, but also indirectly reflect the status and progress of the pig breeding in our country. In order to understand the genetic mechanism of pig growth traits, find SNP which were significantly associated with growth traits, thereby positioning thnctional gene which were associated with growth traits, a GWAS was carried out for two growth traits (backtat thickness and days to 100 kg) of Yorkshire, with the single marker regression method based on linear model and high-density SNP beadchip. In this study, after using FD1R method for multiple testing correction, we found 4 genome-wide significant SNP WU_10.2 15 142776834, WU_10.2 15 142743420, WU_10.2 15 142698412, WU_10.2 15 142730007 which is related to backtat thickness and located on chromosome 15. We found 2 genome-wide significant SNP ALGA0005769, ASGA0004384 which is related to days to 100 kg and located on chromosome 1.Navigate to tunctional genes possibly associated with growth traits. Then the EnseMble online platform SusScroth swine version 10.2 database, will be significant SNPS loci upward downstream extension 1 Mb gene enrichment analysis, positioning to the gene may be associated with growth traits.Among them, with hundreds of kilograms of weight related genes have a nine day age, respectively is SLC19A3, CCL20, DAW1, SPHKAP, RHBDD1, IRS1, COL4A4, TM4SF20, AGFG1; Live backiht related genes have 16, respectively is ONECUT1, ARPP19, MYOSA, MY05C GNBS, BCL2L10, MAPK6, LEO1, TMOD3, SCG3, LYSMD2, TMOD2, DMXL2, GLDN, CYP19A1, TNFAIP8L3. Conclusion, ARPP19 can control mitosis, ONECUT1 has the tunction of promote cell division and growth and development. So the two genes most likely there is tunction relationship and growth traits.
