摘　　要：目的探讨硼酸钠(Borax)对肝癌细胞株Hep G2白细胞介素6(IL-6)表达的影响。方法4. 0 mmol/L的Borax作用Hep G2细胞不同时间点后,实时荧光定量PCR检测细胞中IL-6 mRNA的变化,ELISA法检测细胞培养上清中IL-6的分泌量。将siRNA-MyD88和阴性对照siRNA(siRNA-NC)转染Hep G2细胞24 h后,4. 0 mmol/L Borax作用Hep G2细胞24 h,实时荧光定量PCR探讨Borax上调Hep G2细胞IL-6表达是否经由MyD88途径。结果 4. 0 mmol/L Borax作用5 min后即诱导Hep G2细胞高表达IL-6 mRNA,24 h达高峰。ELISA结果也显示上清液IL-6蛋白24 h达高峰。siRNA-1片段能有效沉默MyD88基因。MyD88基因沉默后,对Borax诱导的Hep G2细胞IL-6的表达上调无影响。结论 Borax经由非My D88依赖途径诱导人肝癌Hep G2细胞表达IL-6。Objective To investigate the effect of Borax on IL-6 expressed in Hep G2 cells. Methods After4. 0 mmol/L Borax treatment in Hep G2 cells at different time points,the expression of IL-6 mRNA was detected by real-time quantitative PCR,and the secretion of IL-6 protein in cell culture supernatant was detected by ELISA. After transfecting siRNA-M yD88 and negative control siRNA（ siRNA-NC） into Hep G2 cell for 24 h and 4. 0 mmol/L Borax was used to treat Hep G2 cells for 24 h,real-time quantitative PCR was used to explore whether Borax raised IL-6 expression in Hep G2 cells via M yD88 pathway. Results After 4. 0 mmol/L Borax action on Hep G2 cell for 5 min,IL-6 mRNA and protein expression began to rise,and was up to peak at 24 th hour. ELISA results also showed that the supernatant IL-6 protein reached a peak at 24 th hour. siRNA-1 fragment could effectively silence the M yD88 gene. M yD88 gene silence does not affect the increase of IL-6 expression in borax-induced Hep G2 cells. Conclusion Borax induces Hep G2 cells express IL-6 via the M yD88 independent pathway.
