摘 要:目的分析溃疡性结肠炎(ulcerative colitis,UC)组织中粒系分化抗原-1(granulocyte differentiation antigen-1,Gr-1)、细胞外调节蛋白激酶(Extracellular signal-regulated kinases,ERK)、脾酪氨酸激酶(Spleen tyrosine kinase,Syk)的表达。方法予以BALB/c小鼠自由饮用5%葡聚糖硫酸钠(dextran sulfact sodium,DSS)诱导UC模型,实验期间观察小鼠大便性状、血便和体重变化。造模第3和7日处死小鼠,取部分结肠固定,HE染色观察病理组织学变化,免疫组化染色方法测定Gr-1、ERK和Syk的阳性表达率。结果造模7日的小鼠体重明显下降,出现血便,病理组织学评分增高。与正常小鼠比较,Gr-1阳性表达率在造模3日小鼠中无明显变化,但在造模7日小鼠明显增高。UC小鼠结肠组织中p-ERK和p-Syk阳性表达率明显高于正常小鼠,且随着病情进展表达水平随之升高。结论急性UC小鼠存在Gr-1、ERK和Syk异常表达,可能是疾病发生发展的原因之一。Objective To investigate the expression of Gr-1,ERK and Syk during the development of ulcerative colitis in mice. Methods BALB/c mice were given 5% DSS to establish acute colitis model. Body weight and hematochezia were observed daily. Mice were sacrificed on the 3 rd and 7 th day of modeling. and part of the colons were fixed Histological examination was performed after hematoxylin and eosin( HE) staining. The expression of Gr-1,ERK and Syk was detected by immunohistochemical method. Results On the 7 th day of modeling,the body weight of the mice decreased significantly,bloody stools appeared,and the histopathological score increased. Compared with normal mice,the positive expression rate of Gr-1 did not change significantly in the mice on the 3 rd day of modeling,but the mice increased significantly on the 7 th day of modeling. The positive expression rates of p-ERK and p-Syk in colon tissue of UC mice were significantly higher than those in normal mice,and the expression level increased with the progression of the disease. Conclusion Abnormal expression of Gr-1,ERK and Syk in acute UC mice may be one of the causes of disease development.
