摘  要:目的探讨酮体对酸敏感通道的影响。方法原代培养大鼠海马神经元,利用膜片钳技术检测乙酰乙酸、β羟丁酸和丙酮三种酮体对海马神经元酸敏感通道开放状态的影响。结果在pH 6.0条件下细胞外液中分别加入乙酰乙酸、β羟丁酸和丙酮,酸敏感通道电流均明显减弱。三种酮体均可显著抑制海马神经元表面酸敏感通道的开放,抑制率分别为92、47和77%。结论酮体可以显著抑制酸敏感通道的开放。生酮饮食治疗难治性癫痫有可能是通过酮体抑制酸敏感通道来实现。Objective To study the effects of ketone bodies on acid-sensing ion channels.Methods In this study the primary hippocampal neurons of rats were selected as the objectives.The effects of acetoacetic acid,β-hydroxybutyric acid and acetone on the open state of acid-sensing ion channels of hippocampal neurons were investigated by patch-clamp technique.Results Under the condition of pH 6.0 the addition of acetoacetic acid,β-hydroxybutyric acid and acetone in the extracellular solution obviously weakened the currents of acidsensing ion channels.All of three ketone bodies could significantly inhibit the opening of the acid-sensing channels on the surface of hippocampal neurons.The inhibition rates were 92%,47% and 77% respectively.Conclusion Ketone bodies could significantly inhibit the opening of acid-sensing ion channels.Ketogenic diet might achieve the treatment of intractable epilepsy through the ketone body inhibition of acid-sensitive ion channels.
