摘  要:【目的】通过研究牛磺酸对急性重型颅脑创伤(traumatic brain injury,TBI)小鼠海马胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)和牛磺酸转运体(taurine transporter,TauT)表达的影响探讨牛磺酸对颅脑机械损伤的治疗作用。【方法】选择BALB/C小鼠18只,随机分为假手术组(Sham组)、脑创伤组(TBI组)、牛磺酸治疗组(Tau组),每组6只。采用液压打击法在小鼠左侧大脑半球制作TBI模型,Sham组只在相同位置开骨窗,不予打击。Sham组、Tau组造模后立刻尾静脉注射牛磺酸(10 mg/kg),TBI组给予相同剂量的无菌生理盐水。脑创伤后24、48 h分别进行免疫组化染色检测伤侧海马TauT和GFAP蛋白表达。【结果】(1)TBI组在脑损伤后24 h较Sham组伤侧海马TauT表达明显下降(P<0.01),与TBI组相比,Tau组小鼠TauT表达明显增加(P<0.01)。伤后48 h,TBI组较Sham组伤侧海马TauT表达下降更加明显(P<0.01),Tau组较TBI组表达上升趋势显著(P<0.01)。(2)伤后24 h,与Sham组相比,TBI组小鼠GFAP表达明显增加(P<0.01);与TBI组相比,Tau组小鼠GFAP表达无显著差异(P=0.198)。伤后48 h,TBI组较Sham组小鼠伤侧GFAP增加趋势更加明显(P<0.01);与TBI组相比,Tau组小鼠GFAP表达明显下降(P<0.01)。【结论】牛磺酸通过上调急性重型脑损伤小鼠脑部海马区牛磺酸转运体、抑制胶质纤维酸性蛋白表达,发挥其对神经组织创伤的治疗作用。[Objective] To explore the therapeutical effect of taurine on the expression of glial fibrillary acidic protein (GFAP) and taurine transporter (TauT) in hippocampus of mice with acute traumatic brain injury (TBI). [ Methods ] A total of 18 BALB/C mice were equally distributed in three groups: Sham group, TBI group and Tau group (TBI plus taurine treatment group), six mice per group. The TBI model was established via lateral fluid percussion injury (FPI) in TBI and Tau groups. Bone window was opened in the Sham group with no brain injury. Mice in the sham and Tau groups were injected with taurine (10 mg/kg) and mice in the TBI group were injected with stroke-physiological saline solution. The expression of TauT and GFAP was examined by immunohistochemistry at 24 h and 48 h after the injury. [ Results ] (1) The number of TauT-positive cells was significantly reduced in the TBI group relative to the Sham group at 24 and 48 h after TBI (P〈0.01). The number of the positive cells immunoreactive to TauT in the Tau group was increased in the percussion injury region after TBI compared with the TBI group at both 24 h and 48 h (P〈0.01). (2) The number of cells immunoreactive to GFAP was enriched in the percussion injury region after TBI compared with Sham group at both 24 h and 48 h (P〈0.01). There was no difference between the treatment groups in the number of cells immunoreactive to GFAP at 24 h (P=0.198), but the number of GFAP immunoreactive astrocytes in the Tau group was significantly fewer than that in the TBI group at 48 h (P〈0.01). [ Conclusion ] Taurinecan upregualte the expression of TauT but suppress the expression of GFAP in hippocampus of mice after TBI. It has a therapeutic effect on nerve tissue trauma.
