摘 要:目的荧光双染法探索牙周炎对肥胖机体脂肪组织间质增生的影响。方法建立饮食诱导型肥胖(饲料诱导16周)与牙周炎(结扎5 d)的复合小鼠模型。DAPI、Di I分别染色睾周脂肪组织的胞核和胞膜,分析肥胖、牙周炎对有核细胞数的效应。结果与苏木素-伊红、甲苯胺蓝染色相比,DAPI-DiI双染可清晰标记脂肪组织的胞核和脂肪细胞轮廓。肥胖显著上调脂肪组织的有核细胞数(P<0.05)。牙周炎对有核细胞数无显著影响(P>0.05)。结论短期牙周感染对肥胖相关的脂肪组织间质增生无明显影响。DAPI-DiI荧光双染有利于脂肪组织学分析包括有核细胞计数。Objective To explore effects of periodontitis on stromal hyperplasia of adipose tissue in obese individuals using DAPI-Dil double-staining. Methods Diet-induced obesity (16 weeks on diet) and periodontitis (5-day ligation) combined mouse models were established. Cell nuclei and membrane in epididymal adipose tissue were stained by DAPI and DiI, respectively, followed by analysis on effects of obesity and periodontitis on nucleated cell number. Results DAPI-DiI double-staining clearly demonstrated cell nuclei and adipocyte outline in adipose tissue compared to hematoxylin and eosin and toluidine blue staining methods. Nucleated cell number was significantly enhanced by obesity (P〈0.05), but not obviously affected by periodontitis (P〉0.05). Conclusion Short-term periodontal infection has no significant impact on obesity-related stromal hyperplasia in adipose tissue. DAPI-DiI double-staining may be helpful for adipose tissue histology including nucleated cell counting.
