摘 要:目的研究阿司匹林对于癫痫大鼠高频振荡(HFO)脑电特性的影响。方法 30只雄性SD大鼠随机等分为癫痫组(E组)和阿司匹林组(A组)。所有大鼠利用脑立体定位仪定位海马(HP)、丘脑(TH)、运动皮质(MC)和前额叶皮质(PC),放置颅内微电极;定位CA3区,放置套管。套管和电极植入术后7d,经套管注射匹鲁卡品2.4μg制作大鼠癫痫模型。癫痫持续状态(SE)后3h,A组腹腔注射阿司匹林20mg·kg-1·d-1,连续给药24d;E组则连续24d腹腔注射等量的生理盐水。SE后4~24 d,对所有大鼠行视频脑电监测,所得数据经由人工判别和小波分析,并对癫痫发作的潜伏期、持续时间、发作频率、HFO发生率及其时空分布进行统计学分析。结果 A组与E组比较,癫痫发作潜伏期显著延长,而癫痫发作频率和持续时间则显著降低(P<0.05);A组总体HFO发生率显著低于E组(P<0.05);E组HFO由HP区最先产生,并广泛传播至TH、MC和PC区;而A组的HFO则主要局限于HP区内。结论阿司匹林通过抑制痫波经由神经网络向海马外的传播扩散发挥其抗癫痫作用。Objective To study the effect of aspirin on electroencephalographic characteristics of high-frequency oscillation (HFO) in epileptic rats. MethodThirty male SD rats were randomly divided into epilepsy group (E group) and aspirin group (A group). All rats were used to locate the hippocampus (HP), thalamus (TH), motor cortex (MC) and prefrontal cortex (PC) using a stereotaxic apparatus. The intracranial microelectrodes were placed; the CA3 area was located and the cannula was placed. Seven days after cannula and electrode implantation, a rat model of epilepsy was produced by cannulation of pilocarpine 2.4μg. Three hours after status epilepticus (SE), group A was given intraperitoneal injection of aspirin 20mg·kg-1·d-1 for 24 days. In group E, the same amount of saline was injected intraperitoneally for 24 days. After 4-24 days of SE, all the rats were monitored by video-EEG. The data obtained were analyzed by hand and wavelet analysis. The statistical analysis of the latency, duration, frequency of seizures, incidence of HFO and spatio-temporal distribution of seizures were performed. Results Compared with group E, the latency of seizures was significantly prolonged in group A, but the frequency and duration of epileptic seizures were significantly decreased (P〈0.05). The incidence of total HFO in group A was significantly lower than that in group E (P〈0.05); HFO in group E which was first generated by the HP region and spread widely to the TH, MC, and PC regions; while the HFO of the A group was mainly confined to the HP region. ConclusionAspirin exerts its antiepileptic effect by inhibiting the transmission of epileptiform waves through the neural network outside the hippocampus.
