摘 要:目的 :探讨对缺血性再灌注脑损伤大鼠采用盐酸托哌酮联合骨髓间充质干细胞移植进行治疗的效果。方法 :将中南大学湘雅医学院实验动物中心提供的48只健康雄性大鼠作为研究对象。将这些研究对象随机分为模型组、干细胞移植组、盐酸托哌酮组及联合治疗组,每组各有12只大鼠。为四组大鼠建立缺血性再灌注脑损伤模型。建模后,对模型组大鼠不使用任何方法进行治疗。对干细胞移植组大鼠采用骨髓间充质干细胞移植进行治疗。对盐酸托哌酮组大鼠采用盐酸托哌酮进行治疗。对联合治疗组大鼠采用盐酸托哌酮联合骨髓间充质干细胞移植进行治疗。然后,比较四组大鼠在接受治疗后的第3 d及第7 d其神经功能缺损的评分、CD34阳性细胞数及脑梗死的体积。结果 :接受治疗后的第3 d及第7 d,与模型组大鼠相比,干细胞移植组大鼠、盐酸托哌酮组大鼠及联合治疗组大鼠神经功能缺损的评分均较低,其CD34阳性细胞数较多,其脑梗死的体积较小(P<0.05)。与接受治疗后的第3d相比,在治疗后的第7d干细胞移植组大鼠、盐酸托哌酮组大鼠及联合治疗组大鼠神经功能缺损的评分均较低,其CD34阳性细胞数较多,其脑梗死的体积较小(P <0.05)。在接受治疗后的第3d及第7d,模型组大鼠神经功能缺损的评分、CD34阳性细胞数、脑梗死的体积相比,差异均无统计学意义(P>0.05)。结论 :对缺血性再灌注脑损伤大鼠采用盐酸托哌酮联合骨髓间充质干细胞移植进行治疗的效果显著,可促进其神经功能的恢复,减小其脑梗死的体积。: Objective To observe the effect of tolperisone hydrochloride cerebral ischemia neurological function recovery and promote angiogenesis after reperfusion. Methods: healthy male SD rats(n=48) were randomly divided into Model group(12,n =6 for each time point),Transplantation group(12,n=6 for each time point), Tolperisone hydrochloride group(12,n =6 for each time point) and Combined treatment group(12,n =6 for each time point, The model of middle cerebrai artery occlusion (MCAO) was made by Longa' s method. The rats of Methadone hydrochloride group, Combined treatment group were given methadone hydrochloride by gavage(l.Sg/kg). To observe the dynamic state changes of neurological deficit, the positive expression of vascular endothelial cell marker CD34 and cerebral infarction volume at different time points. Results: Compared with Model group, neurological scores decreased, cerebral infarction volume reduced and the positive expression of CD34 increased in Combined treatment group(P〈0.05)o Rats after reperfusion, neurological scores decreased, cerebral infarction volume reduced and the positive expression of CD34 increased at 7d than that at 3d(P〈0.05). Conclusion: The result might promote the nerve function recovery after ischemia in rats by Tolperisone hydrochloride combined with stem cell transplantation, and decrease the infarct volume, which mechanism may be related to angiogenesis.
