摘  要:目的探讨卵泡抑素样蛋白5(follistatin-like 5,FSTL5)在不同肝细胞癌(hepatocellular carcinoma,HCC)细胞株中的表达及在HCCLM3细胞迁移、侵袭中的作用。方法取对数生长期HCC细胞(SMMC-7721、HepG2、HCCLM3、MHCC97H、PLC)和永生化正常肝上皮细胞LO2,采用实时荧光定量PCR法检测FSTL5mRNA相对表达量。将对数生长期HCCLM3细胞随机分为转染组和对照组,转染组转染FSTL5的表达质粒,对照组转染空白质粒;转染24h,采用实时荧光定量PCR法检测2组HCCLM3细胞FSTL5mRNA相对表达量,划痕愈合试验检测HCCLM3细胞迁移能力,Transwell小室法检测HCCLM3细胞侵袭能力,三维培养实验检测HCCLM3细胞伪足生长情况。结果FSTL5mRNA相对表达量在SMMC-7721(0.37±0.03)、HepG2(0.43±0.03)、HCCLM3(0.19±0.02)、MHCC97H(0.28±0.02)、PLC细胞(0.35±0.01)均低于LO2细胞(1.04±0.04)(P<0.05),且在HCCLM3细胞表达量最低(P<0.05);转染24h,转染组HCCLM3细胞FSTL5mRNA相对表达量(12.04±1.42)明显高于对照组(1.00±0.25)(P<0.05),划痕愈合率[(24.7±2.4)%]、侵袭细胞数目[(11.7±2.3)%]均明显低于对照组[(86.7±3.7)%、(34.7±5.9)%](P<0.05);三维培养实验结果显示,转染组HCCLM3细胞克隆球未见伪足生成,对照组可见少量伪足生成且延展性较好。结论 HCC细胞(SMMC-7721、HepG2、HCCLM3、MHCC97H、PLC)中FSTL5表达明显降低,过表达FSTL5可抑制HCCLM3细胞迁移和侵袭。Objective To investigate the expression of follistatin-like 5(FSTL5)in different hepatocellular carcinoma(HCC)cell lines and its role in the migration and invasion of HCCLM3.Methods The relative expressions of FSTL5 mRNA in SMMC-7721,HepG2,HCCLM3,MHCC97 Hand PLC cells and immortalized normal liver epithelial cells LO2 in logarithmic growth phase were detected by real-time quantitative PCR.HCCLM3 cells in logarithmic growth phase were randomly divided into transfection group transfected by the over-expressing plasmid of FSTL5 and control group transfected by the blank plasmid.After transfection for 24 h,the relative expressions of FSTL5 mRNA was detected by real time-PCR,the migration ability of HCCLM3 cells was detected by scratch healing test,the invasive ability was detected by Matrigel transwell assay and the growth of pseudopods was detected by three-dimensional culture assay in two groups.Results The relative expression of FSTL5 mRNA was significantly lower in SMMC-7721(0.37±0.03),HepG2(0.43±0.03),HCCLM3(0.19±0.02),MHCC97 H(0.28±0.02)and PLC cells(0.35±0.01)than that in LO2 cells(1.04±0.04)(P0.05),and was the lowest in HCCLM3 cells(P0.05).After transfection for 24 h,the relative expression of FSTL5 mRNA was significantly higher in transfection group(12.04±1.42)than that in control group(1.00±0.25)(P0.05),while the scratch healing rate and the number of cell invasion were significantly lower in transfection group((24.7±2.4)%,(11.7±2.3)%)than those in control group((86.7±3.7)%,(34.7±5.9)%)(P0.05).Three-dimension culture assay showed no pseudopod production of the cloned sphere of HCCLM3 cells in transfection group,but a small amount of pseudopod in a good ductility in control group(P0.05).Conclusion The expression of FSTL5 significantly decreases in SMMC-7721,HepG2,HCCLM3,MHCC97 H and PLC cells,and its overexpression may inhibit the migration and invasion of HCCLM3 cells.
