摘 要:目的研究蒙药乌力地格对胶质瘤生长的影响并初步探讨作用机制。方法建立人胶质瘤细胞系U251裸鼠移植瘤模型,待肿瘤细胞接种后第7天,将裸鼠随机分为3组:对照组、10mg/kg给药组、20mg/kg给药组、40mg/kg给药组。灌胃给予不同剂量的蒙药乌力地格,每隔1天测量肿瘤体积,持续给药14d后处死裸鼠,取出肿瘤,分析蒙药乌力地格对胶质瘤生长的影响。ELISA检测肿瘤组织凋亡情况。qRT-PCR和WB检测不同组肿瘤组织中PI3K、AKT和mTOR的mRNA和蛋白表达水平。WB检测不同组肿瘤组织中自噬相关蛋白LC3-Ⅰ、LC3-Ⅱ和Beclin-1的表达情况。结果蒙药乌力地格呈剂量依赖性的抑制胶质瘤裸鼠移植瘤生长,促进胶质瘤组织中细胞凋亡。蒙药乌力地格呈剂量依赖性抑制PI3K、AKT和mTOR的mRNA和蛋白表达水平。蒙药乌力地格促进胶质瘤细胞自噬。结论蒙药乌力地格抑制胶质瘤生长,其通过PI3K/Atk/mTOR信号通路促进胶质瘤细胞的凋亡,并诱导胶质瘤细胞发生自噬作用。Objective To study the effect of Mongolian medicine Wulidige on the growth of glioma and explore the mechanism.Methods Human glioma cell line U251 was transplanted into nude mice,On the 7 th day after tumor cell inoculation,nude mice were randomly divided into 3 groups:control group,10 mg/kg administration group,20 mg/kg administration group,40 mg./kg administration group.Different doses of Mongolian medicine Wulidige were administered by intragastric administration,and the tumor volume was measured every other day.After 14 days of continuous administration,the nude mice were sacrificed,and the tumor was removed to analyze the effect of Mongolian medicine Wulidige on the growth of glioma.ELISA was used to detect tumor tissue apoptosis.qRT-PCR and WB were used to detect mRNA and protein expression levels of PI3 K,AKT and mTOR in different groups of tumor tissues.WB was used to detect the expression of autophagy-related proteins LC3-I,LC3-II and Beclin-1 in different groups of tumor tissues.Results Mongolian medicine Wulidige dose-dependently inhibits the growth of glioma xenografts in nude mice and promotes apoptosis in glioma tissues.Mongolian medicine Wulidige inhibits the mRNA and protein expression levels of PI3 K,AKT and mTOR in a dose-dependent manner.Wulidige promoted autophagy of glioma cells.ConclusionMongolian medicine Wulidige inhibits the growth of glioma,which promotes apoptosis of glioma cells through PI3 K/Atk/mTOR signaling pathway and induces autophagy in glioma cells.
