摘 要:利用田间试验和EST-SSR标记法分析了皖北30个牡丹栽培品种的遗传多样性。以牡丹品种翠叶紫、白玉兰、雪桂和曹州红的DNA为模板,选择95对EST-SSR引物进行多态性试验,共有61对呈多态性,多态率为64.2%。再用筛选的61对多态性引物分析30个牡丹品种的亲缘关系。结果表明:61对引物在30份牡丹材料中扩增出多态性基因型311个,平均每对引物扩增5.1个;从田间试验获取30个品种的花色、花型和株高等9个表型性状数据;基于分子标记的聚类结果与基于表型性状的聚类结果基本一致,但也有一定的差异。若以整个遗传距离的总平均数为尺度,30个牡丹品种大致可分为5个类群。检测结果显示,由EST-SSR标记和表型性状计算的两个遗传距离矩阵的相关性达到极显著水平。Genetic diversity of 30 Paeonia suffruticosa cultivars in North Anhui Province was analyzed by EST-SSR markers and polymorphic traits. Ninety-five pairs of EST-SSR primers were screened against genomic DNA of cultivars of Cuiyezi, Baiyulan, Xuigui and Caozhouhong, and 61 pairs of primers showed the polymorphism, accounting for 64.2% of total primers. Then 61 pairs of polymorphic EST-SSR primers were used to reveal the genetic relationships among 30 Paeonia suffruticosa cultivars. The results showed that 61 EST-SSR primers had 311 polymorphic loci in 30 Paeonia suffruticosa cultivars, and 5.1 polymorphic loci were detected by each pair of primers averagely. Moreover, the genetic distance based on polymorphic traits, such as flower color, flower type, plant height and other characteristics. The classification of Paeonia suffruticosa ultivars using Unweighted Pair-Group Method with Arithmetic means (UPGMA) based on EST-SSR primers is consistent with the results of phenotypic characteristics, but they also show some differences. With the total average genetic distance as the threshold, 30 Paeonia suffruticosa cultivars could be divided into five groups. Mantel test revealed that there was a significant correlation between the genetic distance matrix calculated by EST-SSR markers and the genetic distance matrix based on the phenotypic traits.
