摘 要:目的 研究急性胰腺炎 (AP)大鼠细胞钙 -镁ATP酶的变化及维生素E对其的影响 ,以进一步探讨维生素E在AP治疗中的作用机制。方法 将 5 6只SD大鼠分为正常组、AP组及AP后维生素E治疗组 3组 ,后两组在制备AP动物模型后分别于 1、5、10h活体取材胰、肝组织 ,通过钙 镁ATP酶的酶组织化学染色检测其活性 ;同时通过逆转录聚合酶链反应 (RT PCR) ,测定其组织钙 镁ATP酶的表达。结果 实验大鼠胰、肝组织细胞钙 镁ATP酶酶组织化学染色阳性率AP组低于正常组 (P <0 .0 5 ) ,且AP组 5h的阳性率低于 1h ;维生素E治疗组的阳性率高于AP组(P <0 .0 5 ) ,且 5h的高于 1h。RT PCR结果显示 :胰腺组织钙 镁ATP酶的表达AP组最低 ,且AP组 5h的表达低于1h ;维生素E治疗组的钙 镁ATP酶表达高于AP组 ,且 5h的表达高于 1h ;肝组织钙 镁ATP酶的表达各组均为阳性 ,各组间无显著性差异。结论 AP时细胞钙 镁ATP酶活性降低 ,可参与AP时细胞钙超载的发生和发展 ,由此引起细胞正常结构与功能的破坏 ,加重AP的病理变化。维生素E通过其抗氧化作用在治疗AP的过程中 ,提高细胞钙 镁ATP酶活性 ,减轻细胞钙超载 。Objective To study the variation of intracellular Ca 2+-Mg 2+-ATPase activity in acute pancreatitis (AP) rats and the effect of VitE so as to study the mechanism of this medication.Methods Fifty-six Sprague-Dawley rats were divided randomly into three groups: normal control group?AP group?treatment group with Vit E. The tissues of pancreas and liver of AP animal model of the last two groups were obtained at 1 sth?5 thh?10 thh and the activity of Ca 2+-Mg 2+-ATPase was studied by using enzyme-histochemistry staining. Meanwhile,the expression of Ca 2+-Mg 2+-ATPase was studied through RT-PCR.Results The results showed that the positive rate of Ca 2+-Mg 2+-ATPase in AP group was lower than that in normal control group by enzyme-histochemistry staining ( P <0.05),and the positive rate of Ca 2+-Mg 2+-ATPase of AP group at 5 thh was lower than that at 1 sth; the positive rate of Ca 2+-Mg 2+-ATPase of treatment group with Vit E and Tet were higher than that in AP group ( P <0.05),and the positive rate of Ca 2+-Mg 2+-ATPase at 5 thh was higher than that at 1 sth. RT-PCR results manifested: Ca 2+-Mg 2+-ATPase expression of groups with Vit E was higher than that in AP group and the expression at 5 thh was higher than that at 1 sth. The expressions of Ca 2+-Mg 2+-ATPase in liver tissues of different groups were all positive without significant difference.Conclusion The activity of intracellular Ca 2+-Mg 2+-ATPase was decreased in AP rats,that might be involved in the occurrence and development of cellular calcium overload,which may lead to the destruction of rnormal cellular structure and function,and exacerbation of pathologic change of AP. VitE which can increase the activity of cell calcium-magnesium ATPase and relieve the cell calcium overload,might play an important role in the treatment of AP in rats.
