乳腺癌中视网膜母细胞瘤基因蛋白的表达及其与p16和c-erbB-2表达的关系

摘　　要：目的　探讨视网膜母细胞瘤基因蛋白 (p Rb)在乳腺癌中的作用及其与 c- erb B- 2和 p16的关系。方法　应用免疫组织化学 S- P法检测 5 9例乳腺癌组织中 p Rb、p16和 c- erb B- 2蛋白的表达状况 ,并观察其与临床病理特征的关系。结果　 p Rb免疫染色主要定位于细胞核 ,其失表达率和低表达率分别为 17%和 2 2 %。 p Rb表达与乳腺癌的组织学类型、分化程度、瘤体大小、淋巴结转移和患者年龄等临床病理特征及 c- erb B- 2表达无相关 (p >0 .0 5 ) ,但与 p16表达相关 (p<0 .0 5 )。结论　乳腺癌中存在广泛的 Rb基因表达的下调或失表达 ,它与 p16的失表达共同参与了乳腺癌的发生。其调控细胞增殖的途径独立于 c- erb B- 2。Objective To investigate the protein expression of retinoblastoma gene (pRb) and its relation with p16 and c-erbB-2 proteins in breast cancer. Methods Immunohistochemical S-P method was used to detect the expression of pRb, p16 and c-erbB-2 proteins in 59 breast carcinomas, their relations with clinicopathologic features were analyzed. Results The low level expression rate and loss expression rate of pRb were 22% and 17%, respectively, with a total reduced expression rate of 39%, which were not significantly associated with the clinicopathologic features, such as histologic type and grading, tumor size, lymphnode involvement, patients' age and c-erbB-2 expression, but significantly related to the loss expression of p16. Conclsions The down regulation or loss expression of pRb is frequently seen in breast carcinoma, which may cooperate with the loss of expression of p16 in carcinogenesis of breast cancer, but its role is independent on c-erbB-2 pathway.
