摘 要:目的 建立血清肝再生增强因子(ALR)的酶联免疫检测方法,了解不同肝病患者血清中ALR 水平及其意义。 方法 利用ALR的酶联免疫方法检测不同肝病患者外周血清中ALR 水平。 结果 所建立的ALR 酶联免疫方法其最低检测到的ALR 浓度为0.1 μg/L,在10 ng/m l范围内抗体与ALR 的反应呈良好的线性关系,与TGF-α,EGF等其它促肝细胞增殖因子无任何交叉反应。重型肝炎患者血清中 ALR 水平最高,急性肝炎次之,两者均非常显著高于正常人,慢性肝炎,肝炎后肝硬变,原发性肝癌和肝外疾病患者(P< 0.05 和P< 0.01);慢性肝炎和肝炎后肝硬变患者血清中ALR 水平亦明显高于正常人(P< 0.05);原发性肝癌和肝外疾病患者血清ALR 水平与正常人无明显差异(P> 0.05)。 结论 ALR 酶联免疫检测方法对于不同肝病患者血清中ALR 的检测具有良好的特异性和敏感性;肝病患者血清 ALRIn order to develop a method of enzyme linked immunoassay(ELISA) for measurement of serum augmenter of liver regeneration(ALR) and to investigate the levels of ALR in serum of patients with various liver diseases. Method: The serum ALR of patients with various diseases was detected by means of ELISA technique. Results: The lowest concentration to be detected by such a technique was 0.1ng purified ALR/ml. The standard curve for ALR revealed that the latter, within the range of 0.1 10.0ng/ml, was linearly related to the antibody and there was no cross reactivity to TGF α, EGF or other hepatocyte proliferative factors. The serum ALR was the highest in severe hepatitis and the second highest in acute hepatitis. The serum ALR in Both the 2 conditions was markedly higher than that in normal subjects, and that in chronic hepatitis, liver cirrohosis, primary hepatocelullar carcinoma and extra-hepatic diseases ( P<0.05 or P<0.01 ); Serum ALR in chronic hepatitis and liver cirrohosis was significantly higher than that in normal subjects ( P<0.05 ). In patients with primary hepatocellular carcinoma and extra-hepatic diseases, serum ALR was not different from that of normal subjects (P>0.05) . Conclusion: The ELISA technique to assay serum ALR was highly specific and sensitive and the increase of serum ALR was strongly related to liver injury and liver regeneration.
