摘 要:研究视网膜脱离后视网膜色素上皮细胞 (retinalpigmentepithelium ,RPE)的增殖情况。利用微穿刺技术将 0 2 5 %Healon注入视网膜下腔造成视网膜脱离 ,并在不同时间 (术后 0 5、1、2、3、4、5、6、7、10、14、2 0、30、6 0天 )摘除眼球。采用免疫组化法 ,观察视网膜脱离后RPE细胞增殖性细胞核抗原(proliferatingcellnuclearantigen ,PCNA)的表达。光镜下确定PCNA染色阳性的RPE并量化增殖反应。结果显示 ,在上述相应各时间点 ,组织切片平均每 0 2 5mm视网膜有PCNA标记的RPE细胞数是 :脱离区分别为 0 0 5 5、0 4 4 4、0 86 1、1 972、3 139、5 833、6 0 2 8、4 917、3 333、2 195、1 0 83、0 195、0 0 5 6 ,非脱离区为 0 0 0 0~ 0 6 39,两组间差异有显著性意义 (P <0 0 5 )。研究表明 ,神经视网膜与RPE细胞分离可诱导RPE细胞PCNA表达。To study proliferation of the retinal pigment epithelium (RPE) after experimental retinal detachment, an antibody that recognizes the specific proliferating cell nuclear antigen (PCNA) in proliferating cells was used. Retinal detachment was produced in cats by subretinal injection of 0 25% solution of Healon through a micropipette. At different intervals the animals were sacrificed and eye globes were fixed in 10% at malin and embeded in paraffin. Histologic sections were processed for immunohistochemisty using an antibody to detect the PCNA protein. Labeled RPE cells were identified, and the proliferative response was quantified. In the retina detached at 0 5, 1, 2, 3, 4, 5, 6, 7, 10, 14, 20, 30, and 60 days, the number of PCNA labled cells was 0 055, 0 444, 0 861, 1 972, 3 139, 5 833, 6 028, 4 917, 3 333, 2 195, 1 083, 0 195, and 0 056 cells per millimeter of retina, respectively. In non detached retina of the same eye, approximately from 0 to 0 639 cells per millimeter of retina were labeled. The difference of PCNA labeled cells between detached and non detached retina of the same eye was significant. These results indicate that RPE cells are induced to proliferate when they lose contact with neural retina.
