p^(16)蛋白在原发性肝细胞癌中的表达及其意义

摘　　要：应用免疫组织化学方法检测了42例原发性肝细胞癌（HCC）和癌旁肝细胞中p16的表达。结果显示，所有癌旁肝细胞p16表达均为阳性，18例癌细胞内p16蛋白阳性，阳性率为42．86％。p16蛋白弥漫性分布在肝细胞浆内，少数细胞核内也可见阳性显色。同一标本中癌旁细胞p16蛋白表达阳性，癌细胞内则可为阴性，表明用免疫组织化学方法在正常细胞中可检测到p16蛋白，而某些癌细胞可因p16缺失或突变等不能表达。p16表达还与HCC的组织学分级有关，分化程度高的HCCp16的表达率为66．67％（12／18），明显高于低分化HCC组（0／4），提示p16失活可能发生在HCC早期。p16 is a new kind of tumor suppressor gene. The expression of p16 protein in 42 cases of primary hepatocellular carcinoma(HCC) and. liver tissues surrounding carcinoma was studied by the method of immunocytochemisty. p16 protein was detected in 18 of 42 case HCC specimens (42. 86% ) and all the liver tissues surrounding carcinoma. The immunocytochemical staining of p16 protein showed in cytoplasma but did not in nuclear. In same specimen,p16 protein was positive in tissues surrounding cancer but may be negative in cancer tissue The expression of p16 was different from that of p53 protein, p" can be detected in normal cells. In some carcinoma p16 protein may be negative because of deletion or mutation of p16 gene The expression of p16 protein also had correlation with histological grades of HCC The expression rate of p16 in high differentiation grade HCC group was obviously higher than that of lower differentiation group The inactivity of p16 may be the early event of HCC.
