摘　　要：目的:观察脓毒症大鼠不同时间血清cTnI、TNF-α和心肌NF-кB的变化,探讨脓毒症大鼠心肌损伤及细胞因子的演变。方法:60只SD大鼠,随机分为两组:对照组、盲肠结扎加穿孔(CLP)组。对照组开腹后不做任何处理,CLP组行盲肠结扎加穿孔。CLP后开始计时。在规定的时点经颈总动脉采血,用酶联免疫吸附试验检测cTnI、TNF-α,同时心肌组织用免疫组化半定量法观察NF-кB的变化。结果:CLP后5,9,12,18 hCLP组血清cTnI均高于对照组(P<0.05或P<0.01),在12 h达到峰值;CLP后2,5,9,12 hCLP组血清TNF-α均高于对照组(P<0.01),随着时间的延长有增加趋势;CLP后各时点CLP组心肌组织NF-кB表达均高于对照组(P<0.01),随着时间的延长有增加趋势。结论:脓毒症时存在心肌损害,致炎性因子表达增高。Objective:The purpose of this study was to investigate the change of cTnI,TNF-α,NF-кB of myocardial in the septic rat.Methods:Male Sprague-Dawley rats were randomly divided into group C,the control group,with non-CLP(rats treated with cecal ligation and perforation to evoke peritonitis);group CLP.Then the rats were killed at different time points,and the level of serum cTnI,TNF-α,the NF-кB of myocardial were measured.Results:The expression of cTnI increased significantly at 5 h,9 h,12 h,18 h after CLP as compared with control group(P<0.05 or P<0.01) and arrived to it′s top at 12 h.The expression of TNF-α increased significantly at 2 h,5 h,9 h,12 h after CLP as compared with control group(P<0.01) and there was an decreasing trend with development of CLP.The expression of NF-кB myocatdial increased significantly in each time as compared with control group(P<0.01),and there was an increasing trend with development of CLP.Conclusion:There exists cardiac injury in septic rats,and inflammatory factor increased.
