摘  要:脂类是构成细胞内外膜质结构的主要成份。在维持膜结构完整性、信息的跨膜传递和调节代谢等方面起着十分重要的作用。恶性肿瘤时生物膜(包括表面膜、亚细胞膜等)的结构和功能均发生明显改变,导致细胞的各种恶性行为,而这些改变必然伴有脂类组成和代谢的改变。但过去对恶性肿瘤中的蛋白质、酶和核酸研究较多,对脂类则少报道。本实验室曾发现用Solt和Farber法诱发肝癌的鼠肝中,蛋白激酶C在癌前期即见明显升高。因磷脂在蛋白激酶C的调节中起关键作用,故研究肝癌中磷脂的代谢将有助于癌变机理的了解。本文系统地研究了二乙基亚硝胺(DEN)诱发大鼠肝癌过程中,胆固醇和磷脂及其各组分含量的动态变化。During the induction of hepatocarcinoma in rat with N-nitrosodiethylamine,the.eontents of protein,total lipid,total cholesterol and total phospholipid were all fluctualedand consisted with the pathological changes of liver. In general, protein and DNA weregradually increased, but total lipid, after reaching a peak level jn 6th week as the pro-tein bo DNA,gradually decreased to the normal level. This was owing to the decreaseof phospholipid 6 weeks after induction, and phospholipid which consisted of 60% oftotal lipid in normal liver. In contrast, total cholesterol including cholesterol andcholesterol ester,which account for only 6% of total lipid,was increased to twice of thenormal value. Among;different phospholipid species, the molar percentage of phospha-tidylcholine (PC) in total phospholipid was gradually decreased, whereas phosphatidyl-ethanolamine (PE) was significantly inoreased before the 8th week of induction andbecame to exceed the amount of PC. from 6th week, then slightly decreased but stillhigher than normal. Phosphatidylinositol(PI)and phosphatidylserine(PS)also decreasedand increased respectively in the late stage of liver carcinogenesis. Cardiolipin (CL)and sphingomyelin(SM) remained relatively unchanged after a greater or smallervariation in the early stage. The significances of the above mentioned changes werediscussed.
