肝复健冲剂对大鼠肝癌C-myc IGF-Ⅱ基因表达的影响

摘　　要：目的　观察中药方剂肝复健冲剂对二乙基亚硝胺(DEN)诱导大鼠肝癌形成过程 C- m yc,IGF- 基因表达的影响 .方法　建立 DEN诱肝癌模型的同时预防性应用肝复健冲剂 ,采用原位杂交方法及计算机图像分析技术检测了大鼠肝癌发生过程中 C- m yc,IGF- 基因的表达变化及肝复健冲剂对其异常表达的影响 .结果　在大鼠肝癌发生过程 C-myc,IGF- 基因阳性信号随着诱癌的进展呈协同增高趋势 ,肝复健冲剂可明显抑制诱癌全过程 C- myc的阳性表达(130 .7± 3.7～ 146 .3± 3.7) ,(4 wk) ;(99.5± 1.9～ 114.6±2 .7) ,(8wk) ;[(84.1± 1.4～ 12 9.3± 1.3) ],(12 wk) ;(80 .1± 2 .5～ 10 0 .6± 7.6 ) ,(16 wk) ;[(76 .1± 0 .8～ 92 .0±3.8) ],(P<0 .0 5 ) .显著抑制 IGF- 阳性表达 (12 5 .4± 6 .9～141.1± 7.5 ) ,(4 wk) ;(95 .7± 8.4～ 116 .1± 5 .6 ) ,(8wk) ;[(10 1.0± 4.5～ 12 2 .1± 1.2 ) ],(12 wk) ;(79.5± 4.8～10 4.5± 4.1) ,(16 wk) ;[(80 .8± 1.5～ 10 1.3± 1.7) ],(P<0 .0 5 ) .结论　肝复健冲剂对 DEN诱导大鼠肝癌的预防作用可能与抑制癌变过程 C- myc,IGF- 等癌基因在肝组织的过量表达有关 .AIM To investigate the effects of Ganfujian Granule preparation on the expression of C-myc, IGF-Ⅱ gene of experimental hepatocarcinoma induced by diethylnitrosamine (DEN) in rats. METHODS Liver carcinogenesis model was established and Ganfujian Granule preparation was used as preventive measure. Effects of Ganfujian preparation on abnormal expression of c-myc and IGF-Ⅱ gene were detected by in situ hybridization technique and analyzed with computer image technique. RESULTS Positive expression of c-myc and IGF-Ⅱ gene increased gradually in the process of rats' hepatocarcinogenesis. Ganfujian granule preparation inhibited the overexpression of two genes strikingly in different stages of hepatocarcinogenesis. Overexpression of c-myc was reduced (130.7±3.7~146.3±3.7), (4 wk); (99.5±1.9~114.6±2.7), (8 wk); [(84.1±1.4 ~129.3±1.3)], (12 wk); (80.1±2.5~100.6±7.6), (16 wk); [(76.1± 0.8~92.0±3.8)] (20 wk) and so did IGF-Ⅱ (125.4±6.9~141.1±7.5), (4 wk); (95.7±8.4 ~116.1±5.6), (8 wk); [(101.0±4.5 ~122.1±1.2)], (12 wk); (79.5±4.8~104.5±4.1), (16 wk); [(80.8±1.5~101.3±1.7)], (P<0.05). CONCLUSION Ganfujian Granule preparation can prevent the occurrence of liver cancer, prolong the survival time by inhibiting the overexpression of c-myc and IGF-Ⅱ oncogenes.
