摘 要:目的 :应用微阵列技术探讨CDC10在星形细胞起源胶质瘤中的表达。方法 :术中收集 17例不同级的新鲜星形细胞起源胶质瘤标本 ,提取总RNA后逆转录成3 3 P标记的cDNA探针 ,与AtlascDNA微阵列杂交 ,获得相应基因表达谱 ,与正常脑组织的基因表达谱比较 ,并应用RT PCR验证结果。结果 :发现CDC10在所有肿瘤基因表达谱中均明显下调 ;经RT PCR验证 ,相对于正常脑组织的高mRNA丰度 ,所有肿瘤中该基因均下调甚至无表达。结论 :应用微阵列平行分析有助于发现已知基因的新功能。CDC10在星形细胞起源胶质瘤中表达下调 。Aim:To explore the expression of CDC10 in human astrocytic gliomas with cDNA array. Methods:17 research samples of different types of astrocytic gliomas were acquired during operation. After isolation of the total RNAs, the 32 P labeled cDNA probes were generated by reverse transcription and then hybridized to the Atlas cDNA array. The gene expression profiles of tumors were obtained, which were then compared to one of the normal brain tissue individually. The result was confirmed with Semi quantitative RT PCR.Results:CDC10 were under expressed in all tumors when compared with the normal brain tissue. It was revealed by RT PCR that the mRNA was dim or not detectable in tumors comparing to the high level in the normal brain tissue. Conclusion:It is helpful to reveal the new function of the known gene to profile gene expressions parallelly by the cDNA array. CDC10 are under expressed in astrocytic gliomas with their real function to be further analyzed.
