摘  要:目的 :评价谷胱甘肽S转移酶π(GST π)和P 糖蛋白 (P glycoprotein ,P gp)在胃癌组织中的表达及其与体外化疗药物敏感性的关系。方法 :对 39例胃癌新鲜手术标本 ,用SABC免疫组化法分别检测GST π和P gp表达 ,同时用MTT法检测原代胃癌细胞对 5 FU、CDDP、ADM和MMC四种化疗药物的体外敏感性 ,以研究其间的关系。结果 :在 39例胃癌标本中GST π和P gp的表达阳性率分别为 66 .67%和 58.97% ;对GST π表达阳性者 ,CDDP和MMC均显示了较强的体外耐药性 ,而对后者表达阳性者 ,ADM显示了较强的体外耐药性。GST π和P gp的高表达与肿瘤细胞对 5 FU的体外耐药性未显示相关关系。结论 :胃癌对CDDP、MMC和ADM等药物的耐药性与其GST π和P gp的高度表达相关 。To study the expression of glutathione S transferase π(GST π) and P glycoprotein (P gp) of human gastric cancer tissues, and their relationship to drug chemosensitivity test in vitro. Methods:The expressions of GST π and P gp in 39 gastric cancer specimens were studied by immunohistochemistry. Simultaneously, the drug chemosensitivity of highly purified tumor cells from gastric cancer tissues to 5 FU, CDDP, ADM and MMC was determined by MTT colorimetric assay. Results: The expressions of GST π and P gp in gastric cancer tissue were 66.67% and 58.97% respectively. The expression of GST π was concordant with the resistance to CDDP and MMC, but not with that of 5 FU and ADM, while the expression of P gp was associated with the resistance to ADM. Conclusions: The expression of GST π and P gp in gastric cancer tissues are closely related to the chemoresistance of MMC, CDDP and ADM in vitro. Detection of GST π and P gp expression, together with MTT chemosensitivity test, is useful for predicting the sensitivity toward various drugs of the gastric cancer tissue under study.
