摘 要:本文对ICM细胞DNA图像定量测量分析技术进行了分析讨论。该技术既不同于显微分光光度术(MSP),也不同于流式细胞光度术(FCM)。其方法是将医学病理上常用的细胞切片、印片或涂片,经福尔马林、甲醇和冰醋酸混合固定剂固定后用Feulgen染色,利用高精度图像采集系统将细胞DNA的显微数字化灰度图像摄入计算机,通过图像处理分析技术对该细胞DNA图像进行定量分析计算,并给出被测细胞DNA的质量、倍体分布及细胞周期等参数。ICM技术方法简便、实用而且准确度高,对人体及其它动物肿瘤或癌症的早期病理分析和诊断研究具有重大参考意义。The new technology of image cytometry (ICM) was presented, analyzed and discussed in this paper. ICM is different from the technology of microspectrophotometry (MSP), and also different from the technology of flow cytometry (FCM). Staining slice of cell fixed by paraformaldehyde, methanol and glacial acetic acid with Feulgen colouring agent, we can acquire digitized DNA microscope gray level images of cell, then store them into computers memory by an advanced image collecting system. With image processing technology, we can analyze and measure the digitized DNA microscope gray level images, obtain the cell DNA mass, their diploid distribution and cell cycle. This method of ICM technology is simple, practical and accurate. And it is very useful in searching the early pathology of the neoplasms in human or animals.
