摘 要:①目的 探讨中波紫外线 (UVB)辐射所致人真皮成纤维细胞凋亡的机制。②方法 建立UVB(辐射强度为 1 .1 76× 1 0 - 4 J/cm2 )对体外培养人真皮成纤维细胞氧化损伤模型。用MTT法检测细胞增殖活性 ,酶法测定胞浆中超氧化物歧化酶 (SOD)、谷胱甘肽过氧化物酶 (GSH px)活性和活性氧 (ROS)的含量 ,流式细胞仪An nexinV FITC法检测细胞的凋亡率和死亡率。③结果 在离体条件下 ,UVB可显著抑制成纤维细胞的增殖活性 ;UVB可降低细胞内SOD ,GSH px活性 ,显著提高ROS的含量 (t =3.87~ 2 4 .4 1 ,P <0 .0 1 ) ;同时可显著提高细胞的凋亡率 (t =57.4 7,P <0 .0 1 )和死亡率 (t =1 3.2 7,P <0 .0 1 )。④结论 UVB所致人真皮成纤维细胞凋亡与其产生氧自由基、抑制细胞清除自由基能力有关。Objective\ To investigate the mechanism of apoptosis of human dermal fibroblasts exposed to ultraviolet B(UVB) in vitro. \ Methods\ An oxidative damage model of human dermal fibroblasts irradiated by UVB (radiated intensity: 1.176 × 10 -4 J/cm 2) was established. The proliferation of fibroblasts was detected with MTT assay. The activity of SOD, GSH px and the amounts of ROS were measured with biochemical methods. The rates of apoptosis and death were detected with flow cytometry (FCM). \ Results\ UVB in vitro, could inhibit the proliferation of fibroblasts and increase the rates of apoptosis ( t= 57.47 , P <0.01) and death ( t=13.27, P < 0.01). The activity of intracellular SOD and GSH px decreased while ROS increased ( t= 3.87 -24.41, P <0.01).\ Conclusion\ Our results indicate that UVB induced apoptosis of fibroblasts is related to its production of oxygen free radicals and inhibition of eradication of free radicals by the cells.
