摘  要:目的 应用微阵列研究少支胶质细胞瘤的基因表达谱。方法 从 2例新鲜少支胶质细胞瘤及 1份正常脑组织标本中提取总 RNA,逆转录为 32 P标记的 c DNA、与 Atlas微阵列杂交 ,洗膜、放射自显影 ,应用专用软件分析所得杂交图。结果 与正常脑组织相比 ,2例少支胶质细胞瘤共有 6 3个基因表达上调 ,4个基因表达下调。 2例肿瘤间基因表达量有显著差异。部分基因的表达趋势与已知基因信息不同。结论 Atlas微阵列可高效显示少支胶质瘤的基因表达谱 ,并为进一步研究肿瘤分子发病机理提供新信息。Objective To study the gene expression profiles of oligodendrogliomas with gene cDNA array. Methods 32 P tagged cDNA probes converted from the total RNA, which had been extracted from 2 fresh samples of oligodendroglioma and 1 of normal brain tissue, were hybridized with the Atlas array. After washing the membranes, the autoradiography was performed and the autoradiograms were analyzed through the special software. Results As compared to the normal brain tissue, there were 63 co upregulated genes and 4 co downregulated genes in these 2 tumor samples. However, a significant quantitative difference existed between them. The expression trend of some genes differed from the known information. Conclusion cDNA array is effective for studying the gene expression profiles of oligodendrogliomas and provides new information for the further research on their molecular mechanisms.
