摘 要:目的 探讨白细胞介素 1(interleukin- 1,IL - 1)基因多态性与新疆地区维吾尔族居民慢性牙周炎 (chronic periodontitis,CP)易感性的关系。方法 收集维吾尔族 4 1例重度 CP患者、4 3例中度 CP患者、4 9例轻度 CP患者和 92名健康对照者的颊粘膜拭子 ,提取 DNA。采用序列特异引物聚合酶链反应 (se-quence specific primers- polymerase chain reaction,SSP- PCR)和聚合酶链反应 -限制性片段长度多态性(polymerase chain reaction- restriction fragment length polymorphism ,PCR- RFL P)方法对其进行 IL - 1A -889/ Nco 和 IL - 1B+395 4 / Taq 位点的基因型测定 ,分析各基因型的分布。结果 IL - 1A- 889/ Nco 基因型在重度 CP、中度 CP、轻度 CP和对照组之间的分布差异无显著性 ;IL- 1B+395 4 / Taq 等位基因 2在重度 CP中的检出率显著高于对照组 ,而在中度 CP、轻度 CP与对照组之间的分布差异无显著性。结论 IL-1B+395 4 / Taq 等位基因 2可能与新疆维吾尔族重度Objective: To investigate the relationship between interleukin-1 (IL-1) gene polymorphisms and the susceptibility of chronic periodontitis in Uighur minority in Xingjiang province of China. Methods: The buccal swabs were collected from 41 severe chronic periodontitis (CP) patients, 43 moderate CP patients, 49 mild CP patients and 92 healthy controls. DNA was extracted from these buccal swabs. Genotypes of the IL-1A-889/Nco I and IL-1B+3954/Taq I were determined by sequence specific primers-polymerase chain reaction (SSP-PCR) and PCR-restriction fragment length polymorphism (PCR-RFLP). Then distribution of genotypes for IL-1A-889 and IL-1B + 3954 were compared among the different groups. Results: (1) There were no significant differences in the distribution of IL-1A-889 among severe CP patients, moderate CP patients, mild CP patients and healthy controls. (2) Frequencies of allele 2 for IL-1B+3954 were higher in severe CP patients than in healthy controls, and the difference was statistically significant. But there were no such significant differences either between moderate CP patients and healthy controls or between mild CP patients and healthy controls. Conclusion: These results suggest that IL-1B + 3953 allele 2 may be a risk indicator for the susceptibility to severe chronic periodontitis in Uighur minority in Xingjiang of China.
