摘  要:目的 :研究非离子表面活性剂对变形链球菌 (S .mutans)致龋基因表达和粘附定植能力的影响。方法 :在培养基中加入非离子表面活性剂 ,分别接种在各致龋基因中融合了编码氯霉素乙酰基转移酶(CAT)的基因片段的S .mutans系列工程菌株 ,通过检测CAT活性 ,观察非离子表面活性剂对S .mutans致龋基因表达的调控作用 ;通过测定S .mutans牢固粘附于玻璃试管的数量 ,观察非离子表面活性剂对S .mutans合成细胞外葡聚糖及形成菌斑能力的影响。结果 :土温 80、土温 4 0可不同程度地增强gtfB、gtfC和gtfD基因的表达 ,聚十八烷乙酯、聚十六烷乙酯可抑制gtfB基因的表达 ,而pac基因的表达则不受非离子表面活性剂的影响。土温 80可促进S .mutans牢固粘附于试管壁 ,聚十八烷乙酯则抑制其粘附。结论 :非离子表面活性剂可在转录水平调节某些致龋基因的表达以及影响其形成菌斑和定植的能力。Objective:To expound the regulative mechanisms of non ionic surfactants on virulence gene expression in Streptococcus mutans and the effects of such substances on the colonization by this microbe.Methods:After addition of surfactants into the culture broth, a series of recombinant strains of S. mutans, in which its virulence genes were fused with the cat gene encoding chloramphenicol acetyltransferase (CAT), were inoculated and cultivated. Through measuring the CAT activities of the samples, the effects of the surfactants on the virulence gene expression were determined; and through measuring the quantities of S. mutans cells firmly attached to glass wall of test tube, the effects of the surfactants on the glucan synthesizing and plaque forming ability by the microbe were also determined.Results:Tween 80 and Tween 40 enhanced the expression of gtfB, gtfC and gtfD, while polystearylether and polycetylether inhibited the expression of gtfB. However, the expression of pac was not affected by Tween 80. Tween 80 facilitated the firm attachment of S. mutans cells to the tube wall, while polystearylether inhibited the process. Conclusion:Non ionic surfactants can regulate the expression of certain virulence genes in S. mutans at transcriptional level and affect the ability of this microbe to form plaque and to colonize.
