摘  要:目的 :探讨阿司匹林对大鼠局灶性脑缺血再灌注损伤肿瘤坏死因子 (TNF)含量及脑组织形态结构的影响。方法 :实验大鼠随机分为阿司匹林组、模型组、假手术组、空白组 ,阿司匹林组连续灌胃给予阿司匹林 ,其他 3组给予生理盐水 ,2 0d后用颈内动脉栓线法造成大鼠大脑中动脉缺血再灌注模型 ,应用放射免疫分析法观察阿司匹林对TNF含量的变化 ,并在光镜和电镜下观察脑组织形态结构的变化。结果 :TNF含量模型组〔(1.6 9± 0 .4 2 8)ng ml〕较假手术组〔(1.32± 0 .4 2 3)ng ml〕明显升高 (P <0 .0 5 ) ,阿司匹林组〔(1.2 6± 0 .111)ng ml〕较模型组显著降低 (P <0 .0 5 ) ;光镜、电镜下观察 ,假手术组神经细胞形态正常 ,模型组神经细胞损伤显著 ,阿司匹林组神经细胞损伤明显减轻。结论 :阿司匹林对大鼠局灶性脑缺血再灌注损伤具有显著的保护作用 ,其机制可能与降低血TNF含量有关。Objective:To study the changes of TNF content in serum and brain morphosis and ultrastructure in rats with focal cerebral ischemia reperfusion injury treated with Aspirin.Methods:Experimental rats were randomly divided into four groups:Aspirin group,model group,sham operative group,contrast group.After exprerimental rats were given Aspirin in Aspirin group and physiological saline in other groups by gastrogavage for 20 days,their middle cerebral artery (MCA) was occluded and recirculated to establish a model of focal cerebral ischemia -reperfusion .Radioimmunoassay was used to measure the level of TNF in serum, and brain morphosis and ultrastructure were observed under light microscope and electron microscope. Results:Serum TNF content〔(1.69±0.428)ng/ml〕 in the model group was marked higher than〔(1.32±0.423)ng/ml〕 in the sham operative group(P<0.05). Serum TNF content〔(1.26±0.111) ng/ml 〕 in Aspirin group was lower than in the model group(P<0.05).Under light microscope and electron microscope, the neurons in the sham operative group were normal while neuron injury in the model group was remarkable,but the injury in Aspirin group was obviously improved.Conclusion:Aspirin had an evident protective effect on focal cerebral ischemia reperfusion injury ,and the effect may be related to the decrease of TNF content in serum .
