摘 要:目的 观察环孢素A(CsA)对体外培养的牛晶状体上皮细胞 (BLEC)增殖的影响。方法 对培养牛晶状体上皮细胞采用3 H TdR掺入法及MTT法 ,研究不同质量浓度CsA( 1~ 16 μg/ml)对体外培养的牛晶状体上皮细胞增殖的影响 ,并在光镜、电镜下进行观察。结果 CsA( 1~ 16 μg/ml)作用 2 4h和 72h对牛晶状体上皮细胞增殖有抑制作用 ,且呈剂量依赖性。 2 4h 5 0 %抑制质量浓度 (IC50 )为 10 37μg/ml,72hIC50 为 6 6 1μg/ml。CsA引起细胞形态学改变的主要特点是细胞胞浆内大量空泡形成 ,且随药物剂量增大 ,细胞形态改变越明显。结论 一定剂量的CsA可抑制体外培养的牛晶状体上皮细胞的增殖 ,为预防后囊混浊提供了新途径。ObjectiveTo investigate the effect of cyclosporin A (CsA) on bovine lens epithelial cells (BLEC) proliferation in culture.MethodsBLECs were cultured in vitro,the 3 rd passage cells were seeded in 96-well plates,and each well was put in 0.1 ml of culture medium.The effects of different concentration of CsA (1~16 μg/ml) on BLEC proliferation were evaluated with 3H-TdR and MTT assaying method at 24 h and 72 h.The morphological changes of BLEC were observed by light microscope and electric microscope.ResultsCsA dosages ranging from 1 μg/ml to 16 μg/ml could inhibite cellular proliferation effectively after incorporated 24 h and 72 h in a dose-dependent manner.The IC 50 of CsA exposed to BLEC for 24 h and 72h was 10.37 μg/ml,6.61 μg/ml by 3H-TdR incorporation,and 11.48 μg/ml,7.06 μg/ml by MTT assaying method respectively.Degenerative cytoplasinic vacuoles were the special changes of BLEC exposed to CsA.ConclusionsCsA at appropriate doses has an effective inhibition on BLEC proliferation.This could be beneficial to the prevention of posterior capsule opacification.
