摘 要:[目的]研究羟基磷灰石(HAP)纳米粒子对肝癌细胞和肝细胞的选择性作用。[方法]以细胞系Bel-7402人肝癌细胞和大鼠肝细胞为对象,倒置显微镜下观察细胞的生长状态,应用活细胞计数法绘制生长曲线。[结果]经HAP纳米粒子(1.4×10-4mol/L)作用的肝癌细胞与其对照组相比,细胞数量逐渐减少,胞质空泡样变,二者存在显著性差异(P<0.01)。而加入不同浓度HAP纳米粒子的肝细胞组,存活细胞数量明显增多,与对照组之间无明显差异。[结论]HAP纳米粒子有明显的体外抗肝癌作用,而对正常肝细胞影响轻微。[Purpose] To investigate the selective effect of hydroxyapatite (HAP) on hepatocarcinoma cells and hepatocyte. [Methods] Research target with cell line Bel-7402 human hepatoma cells and primary rat hepatocyte cells, growth state of cells were observed by inverted microscope, and growth curve were drawed with living cell counting method. [Results]The number of Bel-7402 cells treated by HAP nanoparticles was obviously fewer than control group(P < 0.01), and there was a lot of vacuoles in cytoplasm of Bel-7402 cells. However, the number of survival hepatocyte cells treated by HAP nanoparticles of different concentration were increasing, and had no significant difference than control group. [Conclusion]HAP nanoparticles can inhibit hepatocarcinoma cells in vitro, but have little effect on normal hepatic cells.
