摘　　要：目的:检测DPC4基因在结直肠癌及其癌旁组织中的失活.方法:采用聚合酶链反应(PCR)、单链构象多态性分析(SSCP)检测30例结直肠癌及其癌旁组织的DPC4基因失活情况.结果:30例结直肠癌组织标本中3例出现DPC4基因缺失,4例出现DPC4基因第11位外显子突变;而其癌旁组织则无DPC4基因突变或缺失,有显著性差异.结论:DPC4基因作为一个抑癌基因,参与了结直肠癌的发生.AIM: To investigate the inactivation of DPC4 gene in colorectalcarcinoma.METHODS: Inactivation analysis of the DPC4 gene wereperformed on DNA samples from 30 primary colorectal car-cinomas and 30 tumor-adjacent tissue samples by usingpolymerase chain reaction, sing-strand conformationpolymorphism.RESULTS: Three deletions of DPC4 gene and 4 mutationsof DPC4 gene in exon 11 in 30 cases of colorectal carcinomas;however, no deletions or mutations were detected in 30tumor-adjacent tissue samples.CONCLUSIONS: The DPC4 gene may play a role as a tu-mor-suppressor gene in colorectal carcinoma.
