摘  要:目的 寻求新的HLA A2限制性肿瘤抗原MAGE 2CTL表位。方法 经超基序与量化基序相结合预测的肿瘤抗原MA GE 2的 4个表位作为研究对象 ,标准的51 Cr实验与肽结合实验相结合进行验证。结果 预测表位肽MAGE 2 2 2 0 2 2 8,MAGE 2 2 71 2 79经体外刺激PBMC后可有效的诱导CTL效应产生 ,当效 靶为 10 0∶1时溶破靶细胞效率分别为 74 4% ,68 2 %。HLA A2分子亲和力分析 ,荧光系数分别为 0 61、0 2 4阳性肽荧光系数为 0 79。结论 多肽MAGE 2 2 2 0 2 2 8、MAGE 2 2 71 2 79可作为新的HLA A2限制性CTL表位。Objective To explore and identify novel HLA A2 restricted CTL epitopes of MAGE 2 Methods Four candidate CTL peptides predicted with peptide supermotif and quantitative motif were subjected 51 Cr release assay combined with peptide binding assay were performed to verify these epitopes Results Peptides of MAGE 2 220-228 and MAGE 2 271-279 were confirmed able to elicit CTLs after PBMC stimulation in vitro When effect to target was 100∶1,the lysis percent of target cells were 74 4% and 68 2% respectively This peptide was found to bind with sufficient affinity to the HLA A2 molecule by using a binding assay The fluorescence indexes were 0 61 and 0 24, and positive peptide fluorescence index was 0 79 Conclusion These results indicate that the peptides of MAGE 2 220-228 and MAGE 2 271-279 are novel HLA A2 restricted CTL epitopes
