摘  要:目的 了解重组人生长激素 (r hGH)对肝癌细胞有无促增殖和分化作用。方法 取对数生长期的人肝癌细胞株 74 0 2 ,以不同浓度的r hGH和 (或 )顺铂体外培养 2 4h ,用流式细胞仪测定其细胞增殖周期及细胞凋亡等指标。结果 r hGH组与对照组比较 ,G0 ~G1期细胞数率降低 (P <0 0 5 ) ,S期百分率增高 (P <0 0 5 ) ,顺铂与r hGH合用 ,与单纯顺铂组比较 ,细胞凋亡率增加 (P <0 0 5 )。结论 r hGH在体外作用人肝癌细胞株 74 0 2 ,能诱导细胞分化 ,促进细胞DNA合成 ,减少导致肿瘤复发的静止期细胞 ,提高肿瘤对时相特异性化疗药物的敏感性。与化疗药物同用 。Objective To study the effect of reorganize d human growth hormone (r hGH) on cycle kinetics and apoptosis of liver cancer cell line, 7402 cells. Methods Liver cancer cells (7402 cells) were cultured for 24 h with r hGH at different degrees of concentration with or without DDP. Cells undergoing apoptosis and differentiation were determined by flow cytometry(FCM). Results By comparision of the results between culturing with and without r hGH, percentage of cells in G 0~G 1 phase dropped( P< 0 05), percentage of cells in S phase increased( P< 0 05). Addition of r hGH and DDP to the culture medium increased the percentage of apoptosis of liver cancer cells comparing with those adding DDP only( P< 0 05). Conclusion In vitro r hGH might induce differentiation of liver cancer cells, stimulate the combination of DNA and reduce the cells in G 0~G 1 phase, and it then might improve the sensibility of reaction of tumor to the special staged chemical treatment. Chemotherapy together with r hGH may increase apoptosis of liver cancer cells.
