参附注射液对人白血病细胞生长和凋亡的影响

摘　　要：目的:探讨参附注射液对人白血病HL-60细胞生长抑制和诱导凋亡作用。方法:MTT法检测参附注射液对HL-60细胞生长的抑制作用,流式细胞术分析参附注射液对HL-60细胞凋亡的影响。结果:25 ml/L、50 ml/L、100 ml/L的参附注射液作用48 h,存活率为(87.5±8.02)%、(56.45±5.03)%、(38.38±7.48)%,IC50为68 ml/L。参附注射液50 ml/L作用HL-60孵育48 h后,出现细胞凋亡,G1峰前出现亚二倍体峰。结论:参附注射液对人白血病HL-60细胞生长有抑制作用,其抑制作用与诱导HL-60细胞凋亡有关。Objective: To observe the inhibitory effects of Shenfu Injection and its induction of apoptosis on human leukemia cellsHL-60. Methods: MTT was used to study the inhibitory effects of Shenfu Injection on HL-60 cells growth. The apoptosis rate was analysedby flow cytometry (FCM). Results: MTT showed that HL-60 cells were suppressed after exposed to Shenfu Injection of 25, 50, 100 ml/L,the survival rates were (87.5±8.02)%, (56.45±5.03)%, (38.38±7.48)%, respectively, with IC 50 value of 68 mL/L. Shenfu Injection inducedapoptosis of HL-60 cells and Sub-G1 was showed by flow cytometry. Conclusion: Shenfu Injection can inhibit the growth rates of humanleukemia cells HL-60 and induce its apoptosis.
