摘 要:目的观察辣椒素对人大细胞肺癌NCI-H460细胞线粒体膜电位(Δψm)的影响,探讨其诱导NCI-H460细胞凋亡的线粒体机制。方法体外培养NCI-H460细胞,采用不同终浓度辣椒素进行处理,以不加辣椒素为对照。MTT法检测NCI-H460细胞增殖反应,计算半数抑制率;分别采用流式细胞术Annexin V-FITC/PI法、JC-1活细胞染色、免疫荧光和Western blot检测细胞凋亡、ψm、细胞色素C(Cyt c)的分布和线粒体内Cyt c的表达。结果与对照组比较,辣椒素明显抑制NCI-H460细胞增殖,并诱导NCI-H460细胞凋亡(P<0.05,P<0.01);辣椒素显著降低NCI-H460细胞Δψm、促进Cyt c由线粒体释放至胞浆及降低线粒体内Cyt c的表达(P<0.05)。结论辣椒素对肺癌NCI-H460细胞增殖具有抑制作用,诱导其凋亡,其机制可能是通过线粒体介导的细胞凋亡途径实现。Objective To investigate the effects of capsaicin on mitochondrial membrane potential (Δψm) ofhuman large cell carcinoma NCI-H460; To discuss mitochondrial mechanisms of apoptosis of NCI-H460 cellsinduced by capsaicin. Methods NCI-H460 cells were cultured in vitro and treated with capsaicin at variousconcentrations, and no apsacin-treated group was set as control group. The inhibitory effects of NCI-H460 cellsproliferation and the 50% inhibitory concentration were examined by MTT assay; The effects of capsaicin onNCI-H460 cells apoptosis, Δψm, the distribution of cytochrome C and the expression of cytochrome C in themitochondrial were evaluated by flow cytometry Annexin V-FITC/PI method, JC-1 staining, immunofluorescenceanalysis and Western blot analysis, respectively. Results Compared with the control group, capsaicin significantlyinhibited the proliferation of NCI-H460 cells and induced their apoptosis (P<0.05, P<0.01); Capsaicin significantlyreduced NCI-H460 cells Δψm, promoted cytochrome C which released from mitochondria to cytosol and reduced theexpression of cytochrome C in the mitochondria (P<0.05). Conclusion Capsaicin can inhibit NCI-H460 cellproliferation and induce apoptosis, and the possible mechanisms may be realized through mitochondria-mediatedapoptotic pathway.
