摘  要:目的:观察抑郁症大鼠下丘脑-垂体-肾上腺皮质(HPA)轴功能变化及左归丸对其的作用,探索左归丸治疗抑郁症的作用机理。方法:根据Open-field法筛选60只SPF级SD雄性大鼠,将其随机分为正常对照组、模型组、左归丸组,每组20只动物。应用慢性应激法制作抑郁症大鼠模型。左归丸组大鼠按生药量7.5g·kg-1·d-1灌胃,相当成人(60kg)临床用量的15倍。从应激刺激开始灌胃至应激结束止,共用药21d;ELISA法检测大鼠血清ACTH和血浆CORT的浓度。结果:与正常对照组比较,模型组大鼠血清ACTH和血浆CORT的浓度均显著升高(P<0.05);与模型组比较,左归丸组大鼠血清ACTH和血浆CORT的浓度均显著降低(P<0.05)。结论:抑郁症模型大鼠HPA轴功能亢进,左归丸通过改善HPA轴功能降低大鼠血清ACTH和血浆CORT的浓度,是其治疗抑郁症的机理之一。Objective: The change of HPA axis function of depression model rat and the influence of Zuogui Wan were observed; and action mechanism of Zuogui Wan on depression was explored. Methods: 60 SPF level of SD male rats were selected according to Openfield method, and were randomly divided into the control group, the model group and the Zuogui Wan group, 20 cases for each. Depression rat model was constructed. The Zuogui Wan group received gavage. Serum ACTH and blood plasma CORT concentrations in rats were detected by ELISA method. Results: Compared with the control group, rats’ serum ACTH and blood plasma CORT concentrations in the model group were obviously increased (P<0.05). Compared with the model group, rats’ serum ACTH and blood plasma CORT concentrations in the Zuogui group were obviously decreased (P<0.05). Conclusion: According to improving HPA axis function, rats’ serum ACTH and blood plasma CORT concentrations were decreased and considered to be one of the mechanisms of Zuogui Wan on depression.
