摘 要:目的:探讨全自动银染原位杂交(SISH)方法在乳腺浸润性导管癌组织HER-2基因检测中的应用价值。方法:分别采用免疫组化(IHC)、荧光原位杂交(FISH)、银染原位杂交(SISH)方法对105例乳腺浸润性导管癌组织中的HER-2基因进行检测。结果:HER-2的IHC检测(-)、(1+)、(2+)、(3+)者与FISH检测扩增的符合率分为100%、12.5%、46.5%、89.5%;SISH和FISH检测结果的符合率为100%。结论:SISH是一种快速、准确且重复性高的HER-2基因检测方法,可作为FISH的替代技术。Objective: To evaluate HER - 2 gene expression of invasive breast carcinoma tissues determined bysilver enhanced in situ hybridization(SISH) technique. Methods:Immunohistochemistry (IHC) and fluorescence insitu hybridization( FISH) and SISH was used for HER - 2 gene detection of 105 cases. Results: The coincidencerates of IHC ( - ),(l+),(2 + ),(3 + ) with FISH detection about HER - 2 gene were 100% , 12. 5% ,46. 9% ,89. 5% Respectively. The coincidence rate of SISH with FISH about HER -2 gene amplification were 100%. Conclu-sion: SISH is a rapid, accurate and reproducible detection methods for HER -2 gene. It is a potenial alternative meth-od of FISH.
