摘　　要：目的探讨丝石竹提取物丝石竹皂甙在体外对人胃腺癌细胞株SGC-7901增殖、凋亡的影响及其机制。方法取对数生长期的SGC-7901细胞,加入含不同浓度(20、40、80μg/mL)的丝石竹皂甙分别培养处理24、48、72h,以不含丝石竹皂甙细胞作为对照组。MTT比色法检测细胞增殖活性;流式细胞仪Annexin V/PI双染法检测细胞凋亡;流式细胞仪分析细胞周期;实时荧光定量PCR检测Wnt5a、Wnt8b基因的表达情况。结果 MTT表明,丝石竹皂甙对胃癌细胞株SGC-7901增殖的抑制随处理时间的延长和浓度的增加而增强,其24h的IC50值为40μg/mL;细胞凋亡分析显示,丝石竹皂甙作用24h,细胞凋亡率呈浓度依赖性增加,细胞G0/G1期阻滞,S期细胞数量显著减少;定量PCR结果表明,丝石竹皂甙能下调Wnt5a、Wnt8bmRNA的表达,且呈浓度依赖性。结论丝石竹皂甙通过下调Wnt通路关键基因的表达,抑制胃癌细胞的增殖、促进其凋亡。Objective To investigate the anti-proliferative activity and apoptotic effect of Gypsophila saponins on human gastric adenocarcinoma SGC-7901 cells and its underlying mechanism.Methods SGC-7901 cells at logarithmic growth phase were obtained and cultured with Gypsophila saponins of different concentrations(20,40,80μg/mL)for 24,48 and 72h.Cells cultured without Gypsophila saponins were served as controls.The cell viability was measured by MTT assay,cell apoptosis rate was determined by Annexin V/PI double staining flow cytometry,cell cycle was analyzed by flow cytometry and the expressions of Wnt5 aand Wnt8bmRNA were detected by quantitative RT-PCR.Results MTT assay showed Gypsophila saponins inhibited proliferation of SGC-7901 cells in a time and dose-dependent manner,with IC50 value about 40μg/mL at24 h.Apoptosis analysis revealed apoptotic SGC-7901 cells significantly increased in a dose-dependent manner after treatment with Gypsophila saponins for 24 h.Cell cycle analysis demonstrated Gypsophila saponins treating for 24 harrested most cells at G0/G1 phases and significantly reduced cells at S phase in a dose-dependent manner.Quantitative RT-PCR implied that the expression of Wnt5 aand Wnt8bmRNA was down-regulated in a dose-dependent manner with the increased concentration of Gypsophila saponins.Conclusion Gypsophila saponins inhibit proliferation and promote apoptosis of gastric cancer cells through down-regulation of Wnt genes expression.
