摘　　要：目的检测基因间长链非编码RNA 524346(long intergenic non-coding RNA 524346,LincRNA 524346)在肝细胞癌患者组织(癌、癌旁)及细胞系(正常肝细胞系LO2及肝癌细胞系SMMC-7721、HepG2和Huh7)中的表达情况,探讨LincRNA 524346与肝细胞癌临床病理特征的关系。方法应用实时荧光定量PCR法(real-time quantitative PCR,qRT-PCR)检测LincRNA 524346在61例肝细胞癌患者癌组织、配对癌旁组织以及肝癌细胞系的表达水平,分析其与肝细胞癌患者临床病理特征之间的相关性。用受试者工作曲线(receiver operating characteristics,ROC)评价肝细胞癌组织中LincRNA 524346的表达水平在肝细胞癌早期诊断中的效能。结果 LincRNA 524346在83.6%(51/61)肝细胞癌患者的癌组织中较配对癌旁组织表达下调,差异有统计学意义(P<0.01);LincRNA 524346的表达水平与肝细胞癌患者的病理分期(P=0.021)、肿瘤数目(P=0.013)、是否合并门静脉癌栓(PVTT)(P<0.01)、肝硬化(P=0.005)和微静脉浸润(MVI)(P<0.001)具有显著相关性。结论 LincRNA 524346在肝细胞癌组织中表达降低,其有望作为潜在肝细胞癌早期诊断、转移的分子标志物。Objective To examine the expression of long intergenic non-coding RNA (LincRNA 524346) in hepatocellular carcinoma (HCC) tissues and HCC cell lines, to analyze the relationship between LincRNA 524346 and the clinicopathological status of HCC patients. Methods The differential expressions of LincRNA 524346 among 61 HCC tissues and their paired adjacent non-tumor tissues, in HCC cell lines (SMMC-7721,HepG2 and Huh7) and normal liver cell line (LO2) were detected by quantitative real-time PCR, and their correlation with clinical parameters were analyzed. The sensitivity and specificity of LincRNA expression were analyzed by ROC curves. Results Compared with that in adjacent non-tumor tissues, the expression of LincRNA 524346 in HCC tissues was significantly decreased (P<0.01). Similar results were observed between HCC cell lines and normal liver cell lines. Low expression of LincRNA 524346 was associated with clinical pathological grade (P=0.021), tumor number (P=0.013), complicated with portal vein tumor thrombus (PVTT) (P<0.01), liver cirrhosis (P=0.005) and microvascular invasion (P<0.001). Conclusion LincRNA 524346 may provide a potential biomarker for early diagnosis and evaluation the metastasis of hepatocellular carcinoma.
