摘  要:目的优化鲎试剂动态显色法检测血清样本中脂多糖的处理工艺,降低脂多糖检测中的干扰因素。方法在单因素试验基础上,采用三因素三水平的响应面分析法研究表面活性剂用量、碱用量、处理时间及其交互作用对样本中脂多糖检测结果的影响。结果最佳样本处理工艺为:0.6%Triton X-100与0.020 mol/L KOH等体积混匀作为样本处理液,处理时间10 min。在该工艺下,干扰因素最少,临床符合率可达83.25%。结论建立的最佳样本处理工艺对临床特殊样本(黄疸、溶血、乳糜)的抗干扰能力强,优化后的脂多糖测定方法可显著提高临床样本检测的准确率。Objective To optimize treatment process of chromogenic kinetic limulus amebocyte lysate assay for lipopolysaccharide in serum and reduce interference factors. Methods On the basis of single factor experiments,response surface methodology with 3 factors and 3 levels was used to study the effects and interactions of surfactant,alkali dosage and treatment time. Results The treatment process was optimized as follows:0.6% Triton X-100 mixed with equal volume of 0.020 mol/L KOH as treatment reagent,treatment time 10 min. Under these conditions,the clinical coincidence rate was 83.25%. Conclusions This treatment process can be used to detect special samples,including jaundice,hemolysis and chyle,and can provide a reference for clinical diagnosis.
