宫颈癌中MST1蛋白的表达及临床意义

摘　　要：目的探讨哺乳动物不育系20样激酶1(MST1)在宫颈癌中的表达及其临床意义。方法应用免疫组织化学实验检测139例宫颈癌组织及20例癌旁组织(距离肿瘤≥4 cm)标本中MST1蛋白表达情况,Western blot和q PCR分别检测20例宫颈癌及其配对癌旁组织中MST1蛋白及m RNA表达水平,并分析其表达与临床病理因素及预后之间的关系。结果 MST1主要表达在细胞质。宫颈癌组织中MST1的表达阳性率(27%,38/139)明显低于癌旁组织(80%,16/20),差异有统计学意义(χ~2=21.62,P<0.01),且宫颈癌组织MST1蛋白及m RNA表达水平均明显降低(P<0.01)。Ⅰb+Ⅱa期的宫颈癌患者MST1阳性表达率高于Ⅱb+Ⅳ期,有淋巴结转移者MST1阳性表达率低于无淋巴结转移者(均P<0.05);患者年龄、肿瘤直径、组织学分型及分化程度对MST1阳性表达率无影响。MST1阴性表达患者的生存时间较阳性表达者显著缩短(Log-rank χ~2=28.35,P<0.01)。结论 MST1在宫颈癌中呈低表达,可能是宫颈癌临床治疗和判断预后的新靶点。Objective To investigate the expression and clinical significance of mammalian sterile20-like kinase1(MST1)in cervical cancer.Methods Immunohistochemical method was applied to detect the expression level of MST1protein in specimens of cervical cancer tissues(n=139)and pericarcinomatous tissues(n=20,with≥4cm distance from the primary tumor’s edge).Western blot assay and qPCR were used to detect the protein and mRNA transcription expression levels of MST1in20pairs of cervical cancer tissues and pericarcinomatous tissues,respectively.The correlation betweenMST1expression,clinic pathological features and the prognosis were analyzed.Results MST1was mainly expressed in cytoplasm.The positive expression rate of MST1was significantly lower in cervical cancer tissues(27%,38/139)than that in pericarcinomatous tissues(80%,16/20,χ2=21.62,P<0.01).The expressions levels of MST1protein and mRNA were both lower in the cervical cancer tissues(P<0.01).In cervical cancer,the positive expression rate of MST1inⅠb+Ⅱa stage washigher than that ofⅡb+Ⅳstage(P<0.05),the positive expression rate of MST1in lymph node metastasis was lower thanthat of without lymph node metastasis(P<0.05).Values of age,tumor size,histological type and different iation degreeshowed no significant difference to positive expression rate of MST1.Moreover,the negative expression of MST1displayed a significantly poorer overall survival time than that of positive expression of MST1(Log-rankχ2=28.35,P<0.01).Conclusion MST1shows a lower expression in cervical cancer,which may be a new target for clinical treatment and prognosis of cervical cancer.
