摘  要:目的:观察格雷夫斯病(Graves’ disease,GD)患者外周血树突状细胞(DC)亚群偏移情况及对Th1/Th2失衡的影响。方法:36例初发未治疗GD患者和21例健康志愿者经流式细胞仪检测外周血DC及其亚群mDC/pDC绝对值及比例,分选mDC和pDC,观察其对na?ve T细胞增殖和分化的影响。结果:和健康志愿者相比,GD患者外周血中DC和pDC增多(P<0.05),mDC/pDC比值下降(P<0.05),提示发生向pDC偏移;pDC刺激na?ve T细胞增殖(P<0.01),通过IFN-α诱导na?ve T细胞向Th1分化。结论:DC细胞亚群偏移通过IFN-α诱导Th1分化,是GD发病机制中的重要环节。Objective:To assess the imbalance of dendritic cells(DC)in the peripheral blood from patients with Graves'disease(GD),and the influence on the differentiation of T helpler cells(Th).Methods:In total,36untreated GD patients and21healthy individuals were enrolled in the study.The circulating DC levels,including plasmacytoid DC(pDC)and myeloid DC(mDC),were identified by FACS.In further,pDC and mDC isolated by MACS were co-culture with the na?ve T cells to investigate the role on proliferation and differentiation of Th cells.Results:A significant increase in the percentage of circulating DC and pDC subset was found in GD patients compared with healthy individuals,and GD-derived pDCs were conferred the ability to induce the proliferation of T cells as well as the polarized differentiation of Th1in an IFN-α-dependent manner.Conclusion:The elevated circulating pDCs in GD patients can play a role on the differentiation of Th1in IFN-α-dependent manner,and contribute significantly to the pathogenesis.
