摘 要:目的优化肉豆蔻总木脂素的纯化工艺并对其抗炎活性进行研究。方法采用大孔吸附树脂法富集纯化肉豆蔻总木脂素,以吸附率、解吸率及总木脂素含量为指标,考察树脂型号、洗脱剂浓度、上样量及洗脱剂用量等相关参数;采用二甲苯致小鼠耳肿胀模型,通过测定肿胀度、抑制率考察其抗炎活性。结果总木脂素的纯化采用AB-8型树脂,取粗提物干膏1 g溶解后作为上样液,加入40 m L(含树脂16 g)预处理的树脂,静态吸附12 h,上柱,以2 BV/h流速,先用30%乙醇洗脱至无色,再用70%乙醇300 m L洗脱,收集70%乙醇洗脱液,纯度可达45.8%。肉豆蔻总木脂素高、中、低剂量组的肿胀度与模型组比较,差异均有统计学意义(P<0.05)。结论总木脂素纯化工艺稳定、重复性好,肉豆蔻总木脂素具有明显的抗炎活性。Objective To optimize purification technology of total lignans in Myristicae Semen;To study itsanti-inflammatory effects.Methods Macroporous adsorption resin was used for enrichment and purification of totallignans in Myristicae Semen,with adsorption rate,desorption rate and content of total lignans as indexes.The resintype,concentration of eluent,sample weight and dosage of eluen were investigated.Anti-inflammatory effectswere studied by determining swelling degree and inhibition rate of mice with ear swelling induced by xylene.Results Purification technology of total lignans was processed on type of AB-8resin.1g crude extract was dissolvedas sample;40mL preprocessed resin including16g resin was added and static adsorpted for12hours;30%ethanolwas used firstly to elute to colorless and then300mL70%ethanol was used to elute;the eluent of70%ethanol wascollected.The purity of total lignans was45.8%.There was statistical significance in ear swelling degree of totallignans in Myristicae Semen high-,medium-,and low-dosage groups compared with model group(P<0.05).Conclusion The purification technology of total lignans is stable and reproducible,and total lignans in MyristicaeSemen has significant anti-inflammatory activity.
