摘 要:目的:研究金钗石斛对糖尿病大鼠肝脏胰岛素受体基因表达的影响。方法:将雄性SD大鼠随机分成三组:正常组、糖尿病组、金钗石斛组。糖尿病模型复制成功后各组分别给药8周,测定空腹血糖、血清总胆固醇和甘油三酯的含量。Real-time PCR法和免疫组化法分别分析胰岛素受体基因(Insulin Receptor,InsR)的mRNA表达和蛋白表达。结果:与糖尿病组相比,金钗石斛组大鼠的空腹血糖、血清总胆固醇和甘油三酯分别降低了16.86%、19.79%和14.18%,而肝脏InsR的mRNA表达和蛋白表达分别上调了35.23%和28.57%(P<0.05)。结论:金钗石斛能够改善糖尿病大鼠的胰岛素抵抗,其机制可能与上调大鼠肝细胞InsR的mRNA的表达有关。Objective:To study efficacy of Jinchai Shihu on the expression of insulin receptor gene in liver of diabetic rats.Methods:Male SD rats were randomly divided into the normal group,diabetic group and Jinchai Shihu group.The diabetic models were made,andthe rats were given medicine for8weeks,and the contents of fasting blood glucose,serum total cholesterol and triglyceride were measured.The mRNA expression and protein expression of InsR were analyzed by Real-time PCR and immunohistochemistry.Results:Comparedwith the diabetic group,the fasting blood glucose,serum total cholesterol and triglyceride in the Jinchai Shihu group were reduced by16.86%,19.79%and14.18%respectively,while the expression of InsR mRNA and protein in the liver were increased by35.23%and28.57%(P<0.05).Conclusion:Jinchai Shihu can improve insulin resistance in diabetic rats,and its mechanism may be related to upregulatingthe expression of InsR mRNA in rat hepatocytes.
