翡翠贻贝多糖抑制体外培养肝癌细胞生长的研究

摘　　要：目的:体外实验研究翡翠贻贝多糖(PVPS)对HepG2肝癌细胞生长的影响。方法:将不同浓度的翡翠贻贝多糖作用于HepG2肝癌细胞,用CCK-8法检测细胞增殖情况和细胞贴壁情况,通过划痕实验分析细胞的迁移能力,运用流式细胞术分析细胞周期。结果:作用48h后,低、中、高浓度多糖组H印G2肝癌细胞生长抑制率分别为17.36%、28.46%、96.13%;多糖处理后的H印G2肝癌细胞贴壁率与对照组相比明显减少(P<0.05),且细胞的迁移明显降低(P<0.05);FCM显示细胞周期移行被阻滞于G1期。结论:翡翠贻贝多糖对体外HepG2肝癌细胞增殖有明显的抑制作用,并可降低肿瘤细胞贴壁率和迁移能力,阻滞细胞周期于G1期。Objective:To investigate the effects of PVPS on the growth of hepatocellular carcinoma cells.Methods:Hepatocellularcarcinoma cells were treated with different concentrations of the decoction of PVPS,and its effects on the proliferation and adhesionof hepatocellular carcinoma cells were determined by the CCK-8assay.The scratch test and flow cytometry were used to analyzehepatocellular carcinoma cells'migration and growing cycle.Results:In48hours,in the PVPS group cell growth inhibition ratesrespectively were17.36%,28.46%,96.13%,which measure in low,middle and high concentration.Comparing with the control group,the adherence rate and the rate of Hela cells'migration was reduced significantly(P<0.05).FCM showed that the cell cycle was arrested atG1phase.Conclusion:PVPS can inhibit hepatocellular carcinoma cells in vitro and may be block the growth,conglutination,movementand infiltration of hepatocellular carcinoma cells.
