摘　　要：目的:研究绿原酸(CGA)对β淀粉样蛋白_(25-35)(Aβ_(25-35))诱导的PC12细胞凋亡的保护作用和机制。方法:20μmol/L的Aβ_(25-35)诱导PC12细胞构建细胞损伤模型,并以5个不同浓度CGA干预,CCK-8法检测细胞存活率以确定后续实验CGA的3个浓度。随后,细胞随机分为Control组、Model组及3个浓度CGA组。流式细胞术检测细胞凋亡率;比色法检测丙二醛(MDA)含量、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活力;荧光染色检测线粒体膜电位(MMP);Western blot检测半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)表达。结果:CGA各组较Model组细胞存活率升高、凋亡率下降,SOD和GSH-Px活力升高、MDA水平降低,MMP耗散减少,Caspase-3表达下降(P<0.05)。结论:CGA对Aβ_(25-35)致PC12细胞凋亡有保护作用,其机制可能与改善细胞抗氧化能力和线粒体损伤有关。Objective To study the protective effect of chlorogenic acid(CGA)on the apoptosis of PC12cells induced byβ?amyloid protein23?35(Aβ25?35)and its mechanism.Methods The cells model of death was estab?lished by Aβ25?35(20μmol/L)?induced PC12cells.The cells were interfered with5different concentrations ofCGA.CCK?8assay was used to detect cells viability to determine the3concentrations of CGA in future experi?ments.The cells were divided randomly into control group,model group and interference groups with3differentconcentrations of CGA.Cells apoptosis rates were detected by flow cytometry;colorimetry method was used todetect MDA,SOD and GSH?Px.The mitochondrial membrane potential(MMP)was detected by fluorescent stainingand the expression of caspase?3by western blot.Results Compared with model group,the cells viability of CGAgroups were increased but the apoptosis rates were reduced;the activity of SOD and GSH?Px were increased butthe level of MDA,MMP and caspase?3were decreased(P<0.05).Conclusions CGA has a protective effect onAβ25?35?induced PC12cells apoptosis and it may be related to the improvement of cellular antioxidation capacityand mitochondrial damage.
