摘 要:目的分析缺氧诱导因子(HIF)-1α和幽门螺旋杆菌(Hp)在蒙古族、汉族胃癌组织中的表达,探讨两种因子在蒙、汉两族胃癌发生中的相互作用。方法采用改良的Warthin-Starry(W-S)银染色法检测27例蒙古族和30例汉族胃癌患者Hp感染情况,采用免疫组化(S-P)法、逆转录聚合酶链反应(RT-PCR)法分别检测HIF-1α在癌组织、癌旁组织和正常组织中蛋白和mRNA的表达。分析HIF-1α蛋白和Hp在蒙古族、汉族胃癌组织中表达的关系。结果蒙古族、汉族胃癌患者Hp感染率分别为81.5%(22/27)、66.7%(20/30),差异无统计学意义(χ~2=1.608,P>0.05);HIF-1α蛋白在蒙古族、汉族胃癌患者癌组织中的阳性表达率分别为62.96%(17/27)、70.00%(21/30),mRNA相对表达量分别为0.8914±0.075 0、0.9074±0.0655,两民族间差异无统计学意义;HIF-1α蛋白和mRNA在癌旁和正常组织中均无阳性表达。蒙古族、汉族人群胃癌组织中HIF-1α蛋白的阳性表达与Hp感染均呈正相关(rs分别为0.424、0.617,P<0.05)。结论 HIF-1α和Hp均是胃癌的致病因子,Hp和HIF-1ɑ可能共同促进胃癌的发生。HIF-1α和Hp在蒙古族和汉族胃癌患者中的致病机制可能相同。Objective To detect the expressions of two protein factors hypoxia-inducible factor(HIF)-1αand Helicobacter pylori(Hp)in gastric cancer in Mongolian and Han two nationalities respectively,and to explore the relationship of HIF-1αand Hp in the development of gastric cancer.Methods A modified Warthin-Starry(W-S)silver staining method was used to detect Hp infection in27Mongolian patients with gastric cancer and30Han patients with gastric cancer.Immunohistochemical(S-P)method and reverse transcription polymerase chain reaction(RT-PCR)method were used to detect protein and mRNA expressions of HIF-1αin cancer tissue,paracancerous tissue and normal tissue samples.The relationship of HIF-1αprotein and Hp expression was analyzed between gastric carcinoma tissue samples of Mongolian patient group and Han patient group.Results Hp infection rates were81.5%(22/27)and66.7%(20/30)in Mongolian and Han patients with gastric cancer.The difference in infection rate was not statistically significant between two groups of patients(χ2=1.608,P>0.05).The positive expression levels of HIF-1αprotein were62.96%(17/27)and70.00%(21/30)in gastric cancer tissue samples of Mongolian and Han groups.The relative expression levels of HIF-1αmRNA were0.8914±0.0750and0.9074±0.0655.There was no significant difference in the HIF-1αmRNA expression between the two nationalities.No positive expressions of HIF-1αprotein and mRNA were found in paracancerous tissue and normal tissue samples.There was a positive correlation in the positive expression of HIF-1αprotein and Hp infection in gastric cancer tissue samples of Mongolian and Han patient groups(rs=0.424and0.617,P<0.05).Conclusion HIF-1αand Hp are pathogenic factors of gastric cancer.Hp and HIF-1αmay promote the occurrence of gastric cancer together.The pathogenic mechanism of HIF-1αand Hp in gastric cancer may be the same for Mongolian and Han patients.
