肝龙胶囊联合水飞蓟宾胶囊在体外对大鼠肝星状细胞增殖活性的影响研究

摘　　要：目的研究临床常用抗肝纤维化药物水飞蓟宾胶囊单用及联合肝龙胶囊对大鼠肝星状细胞(HSC)增殖活性的影响,评价肝龙胶囊在体外对水飞蓟宾抗肝纤维化作用的影响。方法采用2因素5水平完全实验,单独或联合使用不同剂量水飞蓟宾胶囊和肝龙胶囊在不同时间作用于HSC,并用MTT比色法测定HSC增殖影响。结果水飞蓟宾胶囊与肝龙胶囊合用增强水飞蓟宾胶囊对HSC的抑制作用,尤其是0.063、0.125和0.250 mg/ml浓度的水飞蓟宾胶囊与不同浓度肝龙胶囊合用后,水飞蓟宾胶囊的作用明显增加,与单用水飞蓟宾胶囊组比较差异均有统计学意义(P<0.05)。水飞蓟宾胶囊联合不同浓度肝龙胶囊的IC50值与单用水飞蓟宾胶囊的IC50值相比较,在24、48和72 h时均有降低,且水飞蓟宾胶囊与不同浓度肝龙胶囊联合后,其合用指数CI<1.0,呈现出协同作用,增强了水飞蓟宾胶囊对HSC细胞的抑制作用。结论肝龙胶囊在体外能增强水飞蓟宾胶囊的抗肝纤维化作用。Objective To investigate the effect of anti-liver fibrosis agent Silibinin Capsules alone or combinedwith Ganlong Capsules on proliferative activity of rat hepatic stellate cells(HSC),and to evaluate the effect ofGanlong Capsules on the in vitro anti-liver fibrosis activity of Silibinin.Methods Two-factors and five-levels fullexperiment was applied for single use or combined use of Silibinin Capsules with different dosages of GanlongCapsules in HSC at different time points.Tetrazolium salt colorimetric assay(MTT)was utilized to determine theeffect of Ganlong Capsules on HSC proliferation.Results Combination of Silibinin Capsules and Ganlong Capsulesenhanced the suppression effect of Silibinin Capsules on HSC proliferation.The effect of Silibinin Capsules wasobviously increased especially at a dosage of0.063mg/ml,0.125mg/ml or0.250mg/ml and in combination withGanlong Capsules,and the differences were significant when compared with the Silibinin Capsule single drug groups(<0.05).Compared with the IC50value of Silibinin Capsules alone,the IC50values of Silibinin Capsules combinedwith Ganlong Capsules at different dosages decreased at24,48and72h.Moreover,the combination indexes(CI)ofthe combined drug therapies were all less than1.0,which illustrated synergistic effect and enhanced the suppressioneffect of Silibinin Capsules on HSC.Conclusions Ganlong Capsules can enhance the anti-liver fibrosiseffect of Silibinin Capsules.
